Biomimetic asymmetric catalysis

Xiao Xiao et al.

Summary: Enzymes are essential for biological transformations and have attracted great attention for their structures and functions. Biomimetic chemistry has rapidly developed in response to the demand for bioactive molecules and efficient synthesis. Biomimetic asymmetric catalysis, inspired by enzymes, has been recognized as one of the most promising strategies for producing valuable chiral compounds. This review summarizes the evolution of biomimetic asymmetric catalysis in terms of biomimetic design, catalyst development, and catalytic transformations, providing insights into bridging the gap between bio-catalysis and chemical synthesis.

Nickel-Catalyzed Asymmetric Hydrogenation of α-Substituted Vinylphosphonates and Diarylvinylphosphine Oxides

Hanlin Wei et al.

Summary: In this study, an efficient earth-abundant transition-metal nickel catalyzed asymmetric hydrogenation of chiral ethylphosphine products was reported. The products were obtained with up to 99% yield, 96% ee (enantiomeric excess) (99% ee, after recrystallization) and 1000 S/C (substrate/catalyst) ratio. This is also the first study on the asymmetric hydrogenation of terminal olefins using a nickel catalyst under a hydrogen atmosphere. Deuterium-labelling experiments and calculations revealed that the two added hydrogen atoms in the products came from hydrogen gas. Additionally, it was suggested that the reaction involves a Ni-II rather than Ni-0 cyclic process based on the weak attractive interactions between the Ni catalyst and terminal olefin substrate.

Manganese catalyzed asymmetric transfer hydrogenation of ketones

Guang-Ya Zhang et al.

Summary: The study investigated the asymmetric transfer hydrogenation of a wide range of ketones catalyzed by manganese complex and chiral PxNy-type ligand under mild conditions. By using 2-propanol as a hydrogen source and combining [MnBr(CO)(5)] with chiral, 22-membered macrocyclic ligand (R,R,R',R')-CyP2N4 (L5) with 2% catalyst loading, various ketones could be enantioselectively hydrogenated to afford highly valuable chiral alcohols with up to 95% ee.

Summary: Asymmetric hydrogenation is an effective method for preparing single enantiomer compounds, but the chemoselective and enantioselective hydrogenation of relatively inert unsaturated groups in substrates with multiple unsaturated bonds remains a challenge. A highly selective hydrogenation method for producing chiral propargylamines and enabling useful chemical transformations has been reported.
