Late-Stage C(sp3)-H Methylation of Drug Molecules

Edna Mao et al.

Summary: Methyl groups are essential in pharmaceutical molecules, and methods to add them to complex molecules are highly desired. This study presents a method for directly introducing methyl groups into saturated heterocycles using decatungstate photocatalysis and nickel-mediated S(H)2 bond formation. The synthetic utility of this method was demonstrated by applying it to a range of drug molecules to produce methylated drug analogues.

Dealkenylative Functionalizations: Conversion of Alkene C(sp3)-C(sp2) Bonds into C(sp3)-X Bonds via Redox-Based Radical Processes

Brady W. Dehnert et al.

Summary: This review summarizes the history and recent advances in dealkenylative functionalization. Radical functionalizations can occur under mild and robust conditions through this deconstructive strategy. The reactions described have high efficiency, good stereoselectivity, tolerate many functional groups, and can be completed within minutes. Rapid diversification of natural products is possible by cleaving the C(sp(3))-C(sp(2)) bond of terpenes and terpenoid-derived precursors.

Investigation of the C-N Bond-Forming Step in a Photoinduced, Copper-Catalyzed Enantioconvergent N-Alkylation: Characterization and Application of a Stabilized Organic Radical as a Mechanistic Probe

Heejun Lee et al.

Summary: Photoinduced, copper-catalyzed couplings of nitrogen nucleophiles with alkyl electrophiles have been shown to be an attractive approach for achieving enantioselective C-N bond formations. This study provides mechanistic insight into a previously reported photoinduced, copper-catalyzed enantioconvergent C-N coupling. Experimental and computational results support the proposed intermediates and their chemical stability and competence in producing the desired C-N coupling product with enantioselectivity.

Metallaphotoredox-enabled deoxygenative arylation of alcohols

Zhe Dong et al.

Summary: Metal-catalysed cross-couplings are widely used for the formation of C-C bonds, but alkyl cross-couplings using alcohols as coupling partners remain relatively underdeveloped. A new method for direct deoxygenative cross-coupling of free alcohols has been developed, showing potential for a wide range of applications, including the pharmaceutical industry. This technology represents a general strategy for combining in situ alcohol activation with transition metal catalysis.

A general N-alkylation platform via copper metallaphotoredox and silyl radical activation of alkyl halides

Nathan W. Dow et al.

Summary: A catalytic union of various functional groups with electronically and sterically diverse alkyl bromides has been achieved through a visible-light-induced metalla-photoredox platform, bypassing traditional high barriers. This regio- and chemoselective protocol is compatible with a wide range of medicinally relevant N-nucleophiles and structurally diverse alkyl bromides. Preliminary mechanistic experiments validate the dual catalytic, open-shell nature of this platform, enabling reactivity previously unattainable in traditional halide-based N-alkylation systems.
