Best practices in metabarcoding of fungi: From experimental design to results

Leho Tedersoo et al.

Summary: The development of high-throughput sequencing technologies has improved our ability to identify fungi and understand their ecological roles. This article provides an overview of the best practices in metabarcoding analysis of fungal communities, covering experimental design, molecular and computational analyses. The study shows that operational taxonomic units are more effective in recovering fungal diversity compared to amplified sequence variants, especially for long markers. Additionally, analyzing the full-length ITS region allows for more accurate taxonomic placement of fungi and other eukaryotes. The authors conclude that metabarcoding analyses of fungi have great potential for integrating fungi into the broader context of microbiome and ecosystem functioning.

Diplodia sapinea in Swedish forest nurseries

Rebecca Larsson et al.

Summary: This study reports the first detection of Diplodia sapinea on diseased seedlings of Pinus sylvestris in two Swedish forest nurseries, highlighting the susceptibility of the seedlings. More research is needed to understand the risk of disease spreading within nurseries and into the forest through infected plant material.

Biological control of emerging forest diseases: How can we move from dreams to reality?

Simone Prospero et al.

Summary: Biological control involves using living natural enemies to control other living organisms, and has gained importance due to increased environmental awareness. However, it is still underutilized in controlling fungal pathogens of forest trees. To enhance success rates, a holistic approach including top-down regulators and bottom-up forces should be adopted.

Seasonal Dynamics of Fungi Associated with Healthy and Diseased Pinus sylvestris Needles in Northern Europe

Ahto Agan et al.

Summary: The relationship between the ecological success of needle pathogens of forest trees and species richness of co-inhabiting endophytic fungi is complex and influenced by various factors such as season, needle age, and specific fungal species composition. The abundance of the most dangerous foliar pathogen of pine, Dothistroma septosporum, is affected by overall fungal richness and specific fungal species composition in Estonia and Norway. The colonization threshold for needle shed in Pinus sylvestris may be breached earlier in Estonia than in Norway, with Lophodermium conigenum playing a significant role in affecting fungal species richness in pine needles.
