Screening and Elucidation of Chemical Structures of Novel Mammalian α-Glucosidase Inhibitors Targeting Anti-Diabetes Drug from Herbals Used by E De Ethnic Tribe in Vietnam

Van Bon Nguyen et al.

Summary: Among indigenous medicinal plants, the MeOH extract of Terminalia triptera Stapf. (TTS) showed the most efficient mammalian alpha-glucosidase inhibition for the first time. Three active compounds, including (-)-epicatechin, eschweilenol C, and gallic acid, were isolated from TTS and determined as novel and potent mammalian alpha-glucosidase inhibitors. In silico studies revealed that these compounds bind to alpha-glucosidase with good binding energy and interact with prominent amino acids. The purified compounds possess anti-diabetic drug properties and low toxicity, making them potential candidates for type 2 diabetes treatment.

Microorganism-Derived Molecules as Enzyme Inhibitors to Target Alzheimer's Diseases Pathways

Thi Hanh Nguyen et al.

Summary: Alzheimer's disease is the most common form of dementia, causing significant impact on individuals, families, and socioeconomics. Current pharmacological therapies for AD target enzymes involved in its pathogenesis. Natural enzyme inhibitors from various sources, particularly microorganisms, show potential for AD treatment. However, there is a lack of comprehensive reviews specifically focusing on enzyme inhibitors from microbial sources. This review addresses this gap by providing an extensive discussion on microbial enzyme inhibitors and their role in the pathogenesis of AD.

Novel Anti-Acetylcholinesterase Effect of Euonymus laxiflorus Champ. Extracts via Experimental and In Silico Studies

Van Bon Nguyen et al.

Summary: This study explored and characterized Euonymus laxiflorus Champ. (ELC) as a natural source of acetylcholinesterase inhibitors (AChEIs) through in vitro and virtual studies. The trunk bark extract of ELC demonstrated significant anti-Alzheimer activity comparable to a commercial AChEI. Multiple compounds with drug properties were identified in the trunk bark extract.

Novel α-Amylase Inhibitor Hemi-Pyocyanin Produced by Microbial Conversion of Chitinous Discards

Thi Hanh Nguyen et al.

Summary: This study discovered a new potential α-amylase inhibitor (aAI) produced by Pseudomonas aeruginosa TUN03 through fermentation. Shrimp heads powder was identified as the most suitable carbon/nitrogen source. Scaling up production in a 14 L bioreactor system yielded a high amount of the aAI in a shorter fermentation time compared to flask fermentation. The major compound, known as hemi-pyocyanin (HPC), exhibited strong inhibitory activity against α-amylase, making it a promising candidate for the development of antidiabetic drugs. Docking studies revealed that HPC binds to the enzyme's active site, inhibiting its activity.

Novel Nematocidal Compounds from Shrimp Shell Wastes Valorized by Bacillus velezensis RB.EK7 against Black Pepper Nematodes

Thi Huyen Trang Trinh et al.

Summary: Among various organic wastes, shrimp shell powder was found to be the most suitable carbon/nitrogen source for producing antinematode compounds via Bacillus veleznesis RB.EK7 fermentation. The study optimized the fermentation process and identified two novel antinematode compounds. These compounds exhibited potential nematicidal effects and showed inhibition of the acetylcholinesterase enzyme.

Sustaining Medicinal Barks: Survival and Bark Regeneration of Amphipterygium adstringens (Anacardiaceae), a Tropical Tree under Experimental Debarking

Leonardo Beltran-Rodriguez et al.

Summary: The study found that bark regeneration rates were highest for female trees harvested in the wet season, and females had higher bark regeneration rates than males. Significant interactions were found between harvest season, harvest treatment, and tree sex diameter on bark regeneration and survival. Wet season and intermediate intensity harvests were shown to promote sound management, but targeting only a single demographic group was not recommended to avoid negative impacts on species demography and bark supply.

Alpha-amylase as molecular target for treatment of diabetes mellitus: A comprehensive review

Navjot Kaur et al.

Summary: Alpha-amylase is a calcium metalloenzyme that aids digestion but also causes postprandial hyperglycaemia, making it a therapeutic target for managing blood glucose levels. Various enzymatic inhibitors have been effective in targeting this enzyme, leading to interest in developing potent alpha-amylase inhibitor molecules. The focus of research has been on designing different derivatives of drug molecules to interact with the target receptor and exhibit biological activities.
