MAD HATTER Correctly Annotates 98% of Small Molecule Tandem Mass Spectra Searching in PubChem

In this paper, a novel computational method called Mad Hatter is presented, which combines results from CSI:FingerID and a structure database to improve annotation accuracy.

Due to the huge chemical diversity, there is no single analysis method that can cover the entire range of metabolites or lipids, leading to the use of different separation methods. This review explores the current use of LC-MS in metabolomics and lipidomics using data from public databases, and highlights potential future trends and improvements needed.

Machine Learning for Absolute Quantification of Unidentified Compounds in Non-Targeted LC/HRMS

This study developed a new method where LC/ESI/HRMS descriptors are used for quantification of compounds even if the structure is unknown. The method showed good accuracy in validation and had smaller errors compared to methods using structural information.

HMDB 5.0: the Human Metabolome Database for 2022

Summary: The Human Metabolome Database (HMDB) has been providing comprehensive information about human metabolites since 2007, and has undergone significant improvements and upgrades in its latest update, HMDB 5.0. These improvements include an increase in the number of metabolite entries, enhancements to metabolite descriptions, new visualization tools, and more accurately predicted spectral data sets. These upgrades are aimed at improving the usability and potential applications of the HMDB in various fields, including human metabolomics, exposomics, lipidomics, nutritional science, biochemistry, and clinical chemistry.

Development of Liquid Chromatographic Retention Index Based on Cocamide Diethanolamine Homologous Series (C(n)-DEA)

Summary: In this study, a system-independent retention index (RI) approach based on CDEA was proposed to index and harmonize retention time (tR) data in liquid chromatography derived under different conditions. The RI values were modeled using techniques like structural barcoding and convolutional neural network. The study addressed the pH effect on elution pattern and the uncertainty associated with the RI model. The proposed RI system enhances identification confidence in suspect and nontarget screening.
