Extracellular Vesicles as Therapeutic Resources in the Clinical Environment

Summary: The native role of extracellular vesicles (EVs) in mediating the transfer of biomolecules between cells has raised the possibility to use them as therapeutic vehicles. The development of therapies based on EVs is now expanding rapidly; here we will describe the current knowledge on different key points regarding the use of EVs in a clinical setting. These points are related to cell sources of EVs, isolation, storage, and delivery methods, as well as modifications to the releasing cells for improved production of EVs. Finally, we will depict the application of EVs therapies in clinical trials, considering the impact of the COVID-19 pandemic on the development of these therapies, pointing out that although it is a promising therapy for human diseases, we are still in the initial phase of its application to patients.

Effects of a 6-wk Sprint Interval Training Protocol at Different Altitudes on Circulating Extracellular Vesicles

Summary: This study investigated the modulation of circulating exosome-like extracellular vesicles (ELVs) after 6 weeks of sprint interval training (SIT) at sea level and different altitudes. The results showed that there were minor differences in the levels of circulating particles, ELV markers, or miRNA between the hypoxic groups. However, the levels of circulating particles increased after training when the hypoxic groups were pooled, and tended to increase at sea level. The miRNA cargo of ELVs seemed to be modulated in hypoxic conditions only.

Effects of an acute exercise bout in hypoxia on extracellular vesicle release in healthy and prediabetic subjects

Summary: The study investigated the effect of acute exercise on plasma concentrations of exosome-like vesicles (ELVs) and markers of multivesicular body (MVB) biogenesis in skeletal muscle. The results showed that acute aerobic exercise increased the plasma abundance of ELVs in healthy subjects under normoxia, but not in prediabetic subjects or under hypoxia. Skeletal muscle analysis suggested a minor role of this tissue in the exercise-induced increase in circulating ELVs.

Summary: This study characterized the protein cargo of extracellular vesicles (EVs) released by adipose tissue (AT) in obese individuals using mass spectrometry analysis. The results revealed that EVs shed by visceral adipose tissue (VAT) in obese individuals contained more inflammation-related proteins compared to those shed by subcutaneous adipose tissue (SAT). Additionally, specific proteins identified in the EVs were associated with disease status and could serve as candidate biomarkers. These findings provide important insights into the mechanisms underlying obesity and related metabolic disorders.

Summary: This study systematically evaluated the effects of blood flow restriction training (BFRT) on anthropometric indicators and blood lipids in overweight/obese adults. The results showed that BFRT significantly reduced BMI, body weight, body fat percentage, and waist circumference. It also decreased total cholesterol and low-density lipoprotein cholesterol levels, lowered triglycerides, and increased high-density lipoprotein cholesterol levels. BFRT can be considered as a safe and effective exercise prescription for personalized weight/fat loss.

Influence of Training and Single Exercise on Leptin Level and Metabolism in Obese Overweight and Normal-Weight Women of Different Age
