MicroRNAs - small RNAs with a big influence on brain excitability

Gareth Morris

Summary: MicroRNAs are short non-coding RNAs that regulate gene expression by targeting complementary sequences in mRNA transcripts, primarily in the 3' untranslated region. They play a role in modulating neural excitability and different processes in the nervous system. Additionally, the expression of microRNAs is regulated by neuronal activity, forming feedback loops.

A complex systems view on the current hypotheses of epilepsy pharmacoresistance

Gabriel Servilha-Menezes et al.

Summary: Drug-resistant epilepsy remains a highly prevalent condition, affecting a significant proportion of patients with epilepsy. The diagnosis and modeling of epilepsy and comorbidities need to adopt a complex system approach. The application of systems biology to epileptology may lead to the discovery of new targets and the development of new antiseizure medications, as well as advancing our understanding of epilepsy and comorbidities.

Posttranscriptional modulation of KCNQ2 gene expression by the miR-106b microRNA family

Kwon-Woo Kim et al.

Summary: Recently emerged miRNAs, specifically the miR-106b family members, have been found to regulate the expression of the KCNQ2 K+ channel protein by binding to its mRNA. The levels of miR-106b decrease while KCNQ2 protein levels increase after birth. Overexpression of miR-106b leads to reduced KCNQ2 protein levels, while inhibition of miR-106b results in increased levels of KCNQ2.

Sodium channelopathies in neurodevelopmental disorders

Miriam H. Meisler et al.

Summary: This review explores the neurological disorders associated with mutations in SCN1A, SCN2A, and SCN8A genes and the therapeutic opportunities arising from recent mechanistic insights. It summarizes the basic elements of sodium channel function used to characterize patient variants and provides examples of the effects of patient mutations on channel function. The review also highlights emerging therapeutic interventions based on new insights into mechanisms of sodium channelopathies.

Summary: Voltage-gated sodium channels play a crucial role in various electrically excitable cells, and mutations in them lead to a wide range of diseases. Mutations affecting periodic paralysis in skeletal muscle were first discovered, providing a foundation for understanding related conditions in the brain.

Opportunities and challenges for microRNA-targeting therapeutics for epilepsy

Gareth Morris et al.

Summary: Epilepsy is a common and serious neurological disorder characterized by recurrent seizures. Current frontline pharmacotherapy with antiseizure drugs is ineffective in 30% of patients. MicroRNA (miRNA) is an emerging therapeutic target that shows promise in treating certain forms of epilepsy with complex underlying pathophysiology, such as temporal lobe epilepsy. Antisense oligonucleotides (ASOs) can inhibit miRNA and have potential for developing therapeutic miRNA-targeting molecules for epilepsy treatment.

Dynamic miRNA changes during the process of epileptogenesis in an infantile and adult-onset model

Petra Bencurova et al.

Summary: The study investigated the miRNA profile of infantile and adult-onset TLE in rats, revealing that infantile-onset TLE leads to changes in the expression of fewer miRNAs and overlaps in dysregulation with some miRNAs in adult-onset TLE. This suggests the potential importance of onset age in the distinct biology underlying TLE.

The Pharmacology and Clinical Efficacy of Antiseizure Medications: From Bromide Salts to Cenobamate and Beyond

Wolfgang Loescher et al.

Summary: Epilepsy is a common and disabling chronic neurological disorder, with antiseizure medications being the main treatment. There are multiple ASMs available for epilepsy therapy and they have diverse mechanisms of action. The goal of epilepsy therapy is to completely eliminate seizures, but this is not achievable in about one-third of patients.
