Teriflunomide as a therapeutic means for myelin repair

Summary: This study demonstrates that teriflunomide modulates oligodendroglial precursor cell homeostasis and promotes myelin repair in the cuprizone mediated de-/remyelination model. Teriflunomide treatment accelerates the generation of oligodendrocytes and restoration of myelin sheaths, and restores mitochondrial integrity within oligodendroglial cells. These findings highlight the importance of de novo pyrimidine synthesis inhibition and mitochondrial homeostasis in successful myelin repair and axonal protection.

Interplay between activation of endogenous retroviruses and inflammation as common pathogenic mechanism in neurological and psychiatric disorders

Summary: Human endogenous retroviruses (ERVs) are retroviral elements integrated into our genome through germline infections and insertions, and have been implicated in the pathophysiology of various disorders, particularly those affecting the central nervous system. Increasing evidence suggests that the induction and expression of ERVs are associated with neurodevelopmental and psychiatric disorders, and a common factor in these disorders is the presence of chronic inflammation. This review discusses the bidirectional relationship between ERV expression and inflammation, and identifies several pathways that contribute to this relationship.

SARS-CoV-2 awakens ancient retroviral genes and the expression of proinflammatory HERV-W envelope protein in COVID-19 patients

Summary: Patients with COVID-19 may exhibit abnormal inflammatory responses, leading to severe disease and long-lasting syndromes. In this study, it was found that exposure to SARS-CoV-2 in vitro can activate the expression of HERV-W proinflammatory envelope protein (ENV) in peripheral blood mononuclear cells, independently of ACE2 receptor and infection. HERV-W ENV was detected in plasma and/or sera of COVID-19 patients, and its levels correlated with ENV expression in T lymphocytes and disease severity. HERV-W ENV was also present in postmortem tissues from various organs of COVID-19 patients. These findings suggest that SARS-CoV-2 can induce the expression of HERV-W envelope protein, and its involvement in the immunopathogenesis of certain COVID-19-associated syndromes emphasizes its relevance in personalized treatment development.

Clonally expanded B cells in multiple sclerosis bind EBV EBNA1 and GlialCAM

Summary: Multiple sclerosis (MS) is an autoimmune disease where autoreactive lymphocytes attack the central nervous system. Epstein-Barr virus (EBV) infection has been linked to MS, with molecular mimicry between EBV nuclear antigen 1 (EBNA1) and central nervous system protein GlialCAM. EBNA1 immunization exacerbates disease in MS mouse models and anti-EBNA1 and anti-GlialCAM antibodies are prevalent in MS patients, providing a mechanistic link between MS and EBV and guiding the development of new MS therapies.

Efficacy and safety of temelimab in multiple sclerosis: Results of a randomized phase 2b and extension study

These trends were sustained over 96 weeks with no safety issues emerging.

The overlap of the transcriptional profile of microglia inflamed in MS with that in other neurodegenerative diseases suggests common mechanisms between primary and secondary neurodegeneration that could benefit from similar therapeutic approaches. The critical role of complement component 1q (C1q) in the activation of microglia inflamed in MS has been identified and potential therapeutic avenues, such as C1q inhibition, have been suggested for addressing chronic white matter inflammation.

A Unique Type of Highly-Activated Microglia Evoking Brain Inflammation via Mif/Cd74 Signaling Axis in Aged Mice

Summary: The study investigated the senescence-associated alterations of microglia in the aged brain at the single cell level, revealing six subgroups of microglia with unique functions. Among them, a unique type of highly activated microglia (HAM) was observed only in aged mice.
