In silico homology modeling, docking and sequence analysis of some bacterial laccases to unravel enzymatic specificity towards lignin biodegradation

Summary: In this study, the amino acid sequences, structural characteristics, and catalytic properties of five bacterial laccases were investigated. Molecular docking and MD simulation results showed that the ligand-bound form of the laccases exhibited higher binding and substrate energy. These findings are important for improving the enzymatic catalysis of bacterial laccases in lignin breakdown and biofuel generation.

Utilization potential of agro-industrial by-products and waste sources: Laccase production in bioreactor with Pichia pastoris

Summary: This study evaluated the production of Madurella mycetomatis laccase from P. pastoris X33 using solid and liquid dextrose from a local corn processing industry and watermelon peel from a local market in a bioreactor. The fermentation strategies involving solid dextrose and liquid dextrose showed significantly higher laccase activity compared to alternative sources and glycerol/methanol. The highest laccase activity was obtained from watermelon peel hydrolysate utilization, despite the lowest cell density in the growth stage. Carbon sources were found to have a significant effect on heterologous protein production in the P. pastoris expression system. These findings demonstrate the suitability of utilizing solid/liquid dextrose and watermelon peel hydrolysate as alternative carbon sources in P. pastoris fermentation for future scale-up.

Characterization of a New Laccase from Vibrio sp. with pH-stability, Salt-tolerance, and Decolorization Ability

Summary: In this study, the laccase coding gene L01 was successfully cloned from a cold-adapted Vibrio sp. LA strain and expressed in the high secretion host Yarrowia lipolytica. The secreted L01 showed high activity and specific activity towards ABTS with the highest activity observed at 35 ℃ and pH conditions ranging from 4.5-10.0. L01 efficiently degraded several synthetic dyes and exhibited robustness, salt tolerance, and pH stability, making it a promising catalytic tool for potential industrial applications.

Selection of a pH- and temperature-stable laccase from Ganoderma australe and its application for bioremediation of textile dyes
