White blood cell count affects fetal fraction and test failure rates in noninvasive prenatal screening

Longwei Qiao et al.

Summary: Increased white blood cell (WBC) count is associated with lower fetal fraction (FF) and higher test failure rates in noninvasive prenatal screening (NIPS). This study found that WBC count negatively correlated with FF and positively correlated with BMI, alanine aminotransferase (ALT), IVF conceptions, and gestational age. High WBC count significantly increased the risk of test failure. These findings highlight the importance of considering WBC count in genetic counseling for pregnant women.

Obstetrical, perinatal, and genetic outcomes associated with nonreportable prenatal cell-free DNA screening results

Mary E. Norton et al.

Summary: The study aimed to assess the outcomes of pregnancies with nonreportable cell-free DNA screening. The results showed that patients with nonreportable results are at higher risk for adverse outcomes, including aneuploidy, preeclampsia, and preterm birth. Therefore, they should be offered diagnostic genetic testing, and clinicians should be aware of the increased risk of pregnancy complications.

Factors Affecting the Fetal Fraction in Noninvasive Prenatal Screening: A Review

Cechuan Deng et al.

Summary: A paradigm shift has occurred in noninvasive prenatal screening with the discovery of cell-free fetal DNA in maternal plasma. Fetal fraction, an important parameter in analyzing screening results, combines biological factors and bioinformatics algorithms. This review aims to provide a deeper understanding of the significance, influencing factors, and interpretation of fetal fraction.

Lipid Metabolism Affects Fetal Fraction and Screen Failures in Non-invasive Prenatal Testing

Jun Cao et al.

Summary: This study found that lipid metabolism is associated with fetal fraction and the rate of screen failures in non-invasive prenatal testing (NIPT). Increased levels of low-density lipoprotein cholesterol and triglycerides were negatively associated with fetal fraction, while high-density lipoprotein cholesterol and the interval between tests were positively correlated with fetal fraction. In addition, high triglyceride levels were independently associated with an increased risk of screen failures.

Clinical impact of additional findings detected by genome-wide non-invasive prenatal testing: Follow-up results of the TRIDENT-2 study

Lisanne van Prooyen Schuurman et al.

Summary: The TRIDENT-2 study examines the clinical impact of genome-wide non-invasive prenatal testing (GW-NIPT) in pregnant women. The study found that additional findings were detected in 0.36% of pregnancies, with most fetal chromosomal abnormalities associated with severe clinical phenotypes. Cases with confined placental mosaicism (CPM) showed increased rates of pre-eclampsia and low birth weight. Maternal findings mainly included malignancies and pathogenic copy number variants.

The Effect of Elevated Alanine Transaminase on Non-invasive Prenatal Screening Failures

Ping Chen et al.

Summary: The study aimed to determine the effects of alanine transaminase (ALT) levels on non-invasive prenatal screening (NIPS) failure rates or no calls due to low fetal fraction (FF). Results showed that increased ALT levels, body mass index, prealbumin, albumin levels, and in vitro fertilization (IVF) conception rates were associated with screening failures. Adjusting these factors may be significant in reducing the probability of no calls due to low FF in NIPS.

Not all low fetal fraction cell-free DNA screening failures are at increased risk for aneuploidy

Samantha Caldwell et al.

Summary: This study found that low fetal fraction (FF) cfDNA failures do not necessarily indicate an increased risk of fetal aneuploidy or adverse pregnancy outcomes. Redrawing may be a suitable next step, as proceeding directly with diagnostic testing for aneuploidy may be unwarranted for most patients.

Association between low fetal fraction in cell-free DNA testing and adverse pregnancy outcome: A systematic review

Peter G. Scheffer et al.

Summary: Low fetal fraction (LFF) in prenatal cell-free DNA (cfDNA) testing may indicate early abnormal placentation and is associated with adverse pregnancy outcomes. Studies have shown associations between LFF and pregnancy-related hypertensive disorders, restricted fetal growth, and preterm birth. Further large prospective cohort studies are needed to explore the relationship between fetal fraction and pregnancy outcomes due to limited available evidence.

Low fetal fraction in cell-free DNA testing is associated with adverse pregnancy outcome: Analysis of a subcohort of the TRIDENT-2 study

Summary: The study found an association between low fetal fraction in prenatal cfDNA testing and adverse pregnancy outcomes, with higher risks of pregnancy-induced hypertension, pre-eclampsia, small for gestational age neonates, and gestational diabetes mellitus in the low fraction group. Further validation is needed to establish the predictive value of low fetal fraction for these outcomes.

Cost-utility of a first-trimester screening strategy versus the standard of care for nulliparous women to prevent pre-term pre-eclampsia in Belgium

Ana Dubon Garcia et al.

Summary: The study assessed the cost-effectiveness of the FMF screening algorithm for pre-term PE in nulliparous women in Belgium. Results showed that the FMF algorithm led to fewer cases of pre-term PE and cost savings compared to the SOC, with similar impact on quality-adjusted life-years. The FMF screening algorithm was cost-saving and more effective in 99.4% of simulations.

Summary: The study compared perinatal mortality rates for small and large-for-gestational-age infants in Europe according to international and national growth charts. The findings suggest that using international charts in Europe could lead to growth restricted infants being reclassified as normal growth and infants with low mortality risks being classified as excessive growth.

The impact of maternal autoimmune disease on cell-free DNA test characteristics
