Construction of a Reduction-responsive DNA Microsphere using a Reduction-cleavable Spacer based on a Nitrobenzene Scaffold

Summary: In this study, a new reduction-cleavable spacer (RCS) based on a nitrobenzene scaffold was designed and synthesized for constructing reduction-responsive oligonucleotides using standard phosphoramidite chemistry. Furthermore, it was demonstrated that the introduction of the RCS in the middle of an oligonucleotide (30 nt) allowed for the construction of a self-assembled microsphere capable of exhibiting a reduction-responsive disassembly.

Design and synthesis of amphiphilic alternating peptides with lower critical solution temperature behaviors

Summary: Amphiphilic peptides with repeating hydrophilic and hydrophobic units were synthesized using Ugi's four-component condensation reaction and their turbidity in aqueous solutions were measured at different temperatures. The study found that the alternating peptides showed thermoresponsive behavior, with peptides containing repeating hydrophilic and hydrophobic units displaying upper critical solution temperature (UCST) behavior, and those with large enough hydrophobic units exhibiting lower critical solution temperature (LCST) behavior. Additionally, thermoresponsive hydrogels were prepared using the alternating peptides as cross-linking points, and their swelling in water was temperature-dependent.

Design of gel-to-sol UCST transition peptides by controlling polypeptide β-sheet nanostructures

Summary: Creating stimulus-responsive materials solely by controlling polypeptide secondary nanostructures is challenging. The synthesized diblock copolymer exhibited gel-to-sol UCST transition behavior in an aqueous solution, with the transition temperature window easily adjusted by changing the copolymer concentration or length of the PBnLT block. The disassembly of initial beta-sheet nanoassemblies caused nanofibril transformation to spherical aggregates with increasing temperature, leading to a gel-to-sol UCST transition. This result inspires a brand-new strategy for the structural design and functional control of materials.
