Reversing the HDAC-inhibitor mediated metabolic escape in MYCN-amplified neuroblastoma

Summary: In MYCN-amplified neuroblastoma, treatment with the HDAC inhibitor vorinostat leads to a reprogramming of the glycolytic pathway, increasing fatty acid oxidation and oxidative phosphorylation. This reprogramming is correlated with reduced levels of MYCN and increased levels of PPARD transcription factors. Additionally, the combination of HDAC and mTORC1 inhibitors increases reactive oxygen species levels. The glycolytic enzymes HK2 and GPI are most affected by this combination. Overall, this combination treatment shows promising results without causing significant toxicity in mice.

Testing hypotheses of a coevolutionary key innovation reveals a complex suite of traits involved in defusing the mustard oil bomb

Summary: Coevolutionary interactions play a crucial role in Earth's biodiversity, and key innovations drive the speciation bursts of species involved. This study focused on the butterfly species Pieris brassicae and found that nitrile specifier protein (NSP) and major allergen (MA) genes are essential for survival on plants containing glucosinolates (GSLs), which are a key innovation in mustard-feeding butterflies. The expression differences of these genes correlated with detoxification performance, particularly when using natural host plants. Positive selection signatures were also detected for NSP and MA genes across Pieris species, highlighting their importance in recent coevolutionary interactions. This research emphasizes the necessity of including ecological relevant experiments to gain insights into coevolutionary dynamics.

The Anticancer Potential of Plant-Derived Nutraceuticals via the Modulation of Gene Expression

Summary: Current studies suggest that diet plays a significant role in cancer-related deaths, with about one-third of deaths being preventable through balanced nutrition. Nutraceuticals derived from plants, such as curcumin and resveratrol, have been shown to have positive effects on gene expression and can reduce the incidence of cancer. These compounds can modulate epigenetic changes and have antioxidant and anticarcinogenic properties. The review also discusses the issue of low bioavailability of nutraceuticals and suggests ways to overcome it for effective oral administration. Understanding the mechanisms by which nutraceuticals influence gene expression could lead to the development of an epigenetic diet for cancer therapy.
