NONHSAG028908.3 sponges miR-34a-5p to promote growth of colorectal cancer via targeting ALDOA

Chuanzhuo Wang et al.

Summary: The aim of this study was to investigate the regulatory mechanism of lncRNA NONHSAG028908.3 on CRC. The results showed that silencing of NONHSAG028908.3 inhibited proliferation, migration, and invasion of CRC cells. Furthermore, NONHSAG028908.3 regulated ALDOA expression by sponging miR-34a-5p, promoting malignant activities in CRC.

The role of circadian clocks in cancer: Mechanisms and clinical implications

Huiwen Liu et al.

Summary: Circadian rhythm is the inherent 24-hour cycle oscillation of biochemical, physiological, and behavioral functions, which is almost universal in eukaryotes. The dysfunction of circadian genes caused by SNPs, deletions, epigenetic modification, and deregulation is strongly associated with cancer risk. This review describes the composition of the circadian rhythm system, highlights the function and mechanism of clock genes in cancer pathogenesis and progression, and discusses their potential clinical implications as prognostic biomarkers and therapeutic targets.

A rationalized definition of general tumor suppressor microRNAs excludes miR-34a

Sophie Mockly et al.

Summary: miR-34a does not meet the definition of tumor suppressor miRNAs, as it is not down-regulated in primary tumors and does not exhibit anti-tumorigenic activity. Therefore, synthetic administration of miR-34a is not efficient against solid tumors.

Effect of 17β-estradiol on the daily pattern of ACE2, ADAM17, TMPRSS2 and estradiol receptor transcription in the lungs and colon of male rats

Iveta Herichova et al.

Summary: The progression of Covid-19 shows sex-dependent features, with a higher survival rate in females possibly attributed to higher levels of 1713-estradiol (E2) compared to males. This study aims to investigate the interaction between E2 signaling and the circadian system in regulating the expression of ACE2 mRNA and related molecules. The results suggest a rhythmic pattern of ACE2 expression in the lungs but not in the colon, influenced by E2 treatment. The findings highlight the importance of considering the daily pattern of SARS-CoV2 entry pathway components and their responsiveness to E2 in the timing of Covid-19 pharmacological therapy.

2.4 GHz Electromagnetic Field Influences the Response of the Circadian Oscillator in the Colorectal Cancer Cell Line DLD1 to miR-34a-Mediated Regulation

Sona Olejarova et al.

Summary: This study found that radiofrequency electromagnetic fields (RF-EMF) had a significant impact on the regulation of the peripheral circadian oscillator mediated by miR-34a in colorectal cancer cells. The inhibitory effects of miR-34a on clock gene expression, cell proliferation, and migration were weakened by exposure to RF-EMF.

Period 2 Suppresses the Malignant Cellular Behaviors of Colorectal Cancer Through the Epithelial-Mesenchymal Transformation Process

Summary: The expression of PER2 is decreased in colorectal cancer, and knocking down PER2 leads to the acquisition of malignant features in colorectal cancer cells. PER2 knockdown may activate the Snail/Slug axis through inhibiting p53, thereby promoting the activation of the epithelial-mesenchymal transition (EMT) pathway.

A promising effect of zerumbone with improved anti-tumor-promoting inflammation activity of miR-34a in colorectal cancer cell lines

Razieh Dehghan et al.

Summary: The cross-talk between inflammation and colorectal cancer cells is mainly mediated by a complex of cytokines, chemokines, and growth factors. MicroRNA, particularly miR-34a, plays a vital role in regulating tumor suppression. Combination therapy using miRNA-based agents and chemotherapeutic drugs has been shown to increase drug sensitivity and enhance the antitumor effect. The anti-inflammatory activity of Zerumbone (ZER) has been investigated in various cancers, showing significant suppression of inflammatory cytokines. The combination of miR-34a and ZER demonstrated promising results in inhibiting cytokine expression and reducing migration and proliferation of CRC cells. Additionally, a compensatory effect between different therapeutic agents was observed in cases of drug resistance.

The role of circadian genes in the pathogenesis of colorectal cancer

Saiedeh Razi Soofiyani et al.

Summary: Colorectal cancer is the third most common cancer in humans, with unclear development mechanisms but known associations with genetic, environmental, and epigenetic risk factors. The circadian rhythm, controlled by circadian clock genes, cytokines, and hormones, regulates various biological processes and disruptions in this rhythm have been linked to diseases such as cancer. The pathways of circadian genes in the pathogenesis of CRC are still underexplored, but understanding them may lead to new therapeutic approaches.

Post-transcriptional modulators and mediators of the circadian clock

Summary: The endogenous circadian timekeeping system drives 24-h rhythms in gene expression and coordinates physiology, metabolism, and behavior in organisms. Regulation at various levels is crucial for its accurate functioning, including a core oscillatory feedback loop mediated by transcription and translation. Post-transcriptional regulation also plays a key role in shaping the rhythmicity of mRNAs and clock proteins accumulation.
