Cellular crosstalk of glomerular endothelial cells and podocytes in diabetic kidney disease

Shan Jiang et al.

Summary: Diabetic kidney disease (DKD) is a serious complication of diabetes that leads to end-stage renal disease (ESRD). Damage to glomerular endothelial cells and podocytes is an early feature of DKD. Recent studies have highlighted the importance of cellular crosstalk in DKD progression, although the underlying pathways are complex and require further investigation.

Honokiol improves endothelial function in type 2 diabetic rats via alleviating oxidative stress and insulin resistance

An He et al.

Summary: This study aimed to investigate the effect of Honokiol (HKL) on endothelial dysfunction in type 2 diabetic rats and its potential mechanism. The results demonstrated that HKL could ameliorate endothelial dysfunction, alleviate insulin resistance, and reduce oxidative stress, suggesting that HKL may be a promising treatment option for diabetes.

Hypoxia, acidification and oxidative stress in cells cultured at large distances from an oxygen source

Natali D'Aiuto et al.

Summary: The interplay between hypoxia and oxidative stress is important for understanding the effects of hypoxic microenvironments on cells. The study found that high accumulation of CO2 contributes to acidification in hypoxic conditions. Modestly hypoxic cells close to the oxygen source showed increased oxidative stress, while cells at large distances from the oxygen source displayed higher levels of hypoxia and milder oxidative stress.

Sirtuin 3 Deficiency Aggravates Kidney Disease in Response to High-Fat Diet through Lipotoxicity-Induced Mitochondrial Damage

Monica Locatelli et al.

Summary: This study found that Sirt3 deficiency makes mice more susceptible to damage from high-fat diets, resulting in more severe kidney diseases. This suggests that protecting mitochondria may be a method to prevent high-fat diet-induced renal injury.

Angiopoietin-2 inhibition attenuates kidney fibrosis by hindering chemokine C-C motif ligand 2 expression and apoptosis of endothelial cells

Fan-Chi Chang et al.

Summary: Plasma levels of angiopoietin-2 are elevated in patients with chronic kidney disease (CKD), and this increase is associated with the development of kidney failure. Overexpression of angiopoietin-1 in mouse models of progressive kidney disease attenuates macrophage infiltration, endothelial cell apoptosis, microvascular rarefaction, and fibrosis. Additionally, an angiopoietin-2 inhibitor has inhibitory effects on macrophage infiltration, microvascular rarefaction, and fibrosis in these models.

Low Nephron Number Induced by Maternal Protein Restriction Is Prevented by Nicotinamide Riboside Supplementation Depending on Sirtuin 3 Activation

Anna Pezzotta et al.

Summary: Reduced nephron number at birth, associated with critical gestational conditions, may increase the risk of hypertension and chronic kidney disease in adulthood. This study investigated the potential of stimulating SIRT3 expression through nicotinamide riboside (NR) supplementation to improve nephron number in a murine model on a low protein (LP) diet. The findings showed that NR supplementation restored SIRT3 expression, improved glomerular podocyte density and preserved renal capillaries, possibly through activation of PGC-1 alpha. Furthermore, NR restored SIRT3 activity, resulting in improved mitochondrial morphology and protection against oxidative damage. These results suggest that boosting SIRT3 during nephrogenesis may prevent nephron mass shortage at birth.

Post-translational modifications by SIRT3 de-2-hydroxyisobutyrylase activity regulate glycolysis and enable nephrogenesis

Luca Perico et al.

Summary: The study reveals a novel role for the NAD(+)-dependent deacylase SIRT3 in kidney development, controlling early renal development through the regulation of epigenetics and metabolic processes. Deficiency of SIRT3 leads to decreased nephron progenitors, impaired nephrogenesis, and a reduced number of nephrons in the developing kidney.
