Depicting environmental gradients from Malaise trap samples: Is ethanol-based DNA metabarcoding enough?

Caroline Chimeno et al.

Summary: DNA metabarcoding is revolutionizing biodiversity research by offering a holistic taxonomic approach. It is important to determine the effectiveness of DNA metabarcoding and compare it with tissue DNA in order to conserve ecological and environmental information. Ethanol can be used as a preservative for DNA and can retain this information without damaging the samples.

Facing the infinity: tackling large samples of challenging Chironomidae (Diptera) with an integrative approach

Caroline Chimeno et al.

Summary: In this study, integrative taxonomy was applied to the diverse fly taxon Chironomidae (Diptera) using a three-level subsampling method and morphological and molecular identification methods. The results showed that identifying less than 10% of a sample's contents can reliably detect over 90% of its species diversity. However, mistakes were made due to the large amount of material, and molecular methods could not provide species information in some cases. Therefore, an integrative approach is crucial when studying non-biting midges in ecological frameworks.

Convergence of dominance and neglect in flying insect diversity

Amrita Srivathsan et al.

Summary: Using DNA barcoding, the authors analyzed the diversity of flying insects and found that more than half of the local species diversity is represented by only 20 insect families, most of which have been neglected in terms of taxonomy. The dominance of these families in insect diversity remains consistent across various factors such as clade age, continent, climatic region, and habitat type. These families, however, suffer from taxonomic neglect and urgent measures are needed to address this issue in biodiversity science.

Peering into the Darkness: DNA Barcoding Reveals Surprisingly High Diversity of Unknown Species of Diptera (Insecta) in Germany

Caroline Chimeno et al.

Summary: This study estimates the species richness of four challenging groups of flies in Germany and finds that there are at least 1800-2200 species awaiting discovery and description. This suggests a higher proportion of undetected biodiversity in Germany than previously thought. Further research is needed to determine the size of the German insect fauna, particularly in taxonomically challenging families of flies and wasps.

A re-analysis of the data in Sharkey et al.'s (2021) minimalist revision reveals that BINs do not deserve names, but BOLD Systems needs a stronger commitment to open science

Rudolf Meier et al.

Summary: Halting biodiversity decline is crucial but hindered by taxonomic impediments, including undescribed and superficially described species. A recent study attempted to address the issue by using COI barcode clusters for braconid wasps in Costa Rica, but the accuracy of the descriptions is questionable.

Emerging technologies revolutionise insect ecology and monitoring

Roel van Klink et al.

Summary: Recent technological advances have the potential to revolutionise insect ecology and monitoring by providing unprecedented possibilities for studying insects. Technologies such as computer vision, acoustic monitoring, radar, and molecular methods offer advantages but also require collaboration to establish international standards.

A quick evaluation of ecological restoration based on arthropod communities and trophic guilds in an urban ecological preserve in Mexico City

Summary: This study compared the diversity and functional traits of arthropods in restored sites with conserved and disturbed sites in the Pedregal de San Angel Ecological Reserve. The results showed that restoration efforts have not yet successfully differentiated restored sites from disturbed sites.

ONTbarcoder and MinION barcodes aid biodiversity discovery and identification by everyone, for everyone

Amrita Srivathsan et al.

Summary: This study introduces a cost-effective and simple DNA barcoding method that can be used in various settings. The research provides amplicon coverage recommendations based on multiple runs of the latest generation of MinION flow cells, and introduces a novel software to address bioinformatics challenges related to MinION reads. Additionally, the study suggests that MinION barcodes are highly similar to Sanger and Illumina barcodes, and that MinION technology has improved rapidly since 2018.
