Construction and biological characterization of an infectious full-length cDNA clone of a Chinese isolate of Wheat yellow mosaic virus

Summary: In this study, infectious cDNA clones of Wheat yellow mosaic virus were constructed for versatile usage. An Agrobacterium-mediated inoculation system allowed WYMV to infect Nicotiana benthamiana leaves, while in vitro transcripts using the SP6 promoter promoted systemic infection in wheat plants. The optimal temperature for virus multiplication and systemic infection was found to be 8 degrees Celsius, and a synergistic effect with Chinese wheat mosaic virus was detected.

Geminivirus V2 protein disrupts the calmodulin-CAMTA3 interaction to counteract RNAi defense, showing the versatility of host antiviral defense and viral counter-defense.

Summary: By utilizing a potato virus X (PVX)-based expression vector, the expression mechanism of P3N-PIPO in turnip mosaic virus and wheat yellow mosaic virus was successfully investigated. Mass spectrometry identified the peptides spanning the slippery site of P3N-PIPO for the first time, providing direct evidence of the frameshift expression. Mutagenesis analysis revealed the importance of the Hexa-adenosine site for frameshift expression. The PVX-based expression vector proved to be a valuable tool for studying the expression mechanism of P3N-PIPO in Potyviridae.
