Summary: This study investigated the efficacy of OCT4&SOX2 specific cytotoxic T lymphocytes (CTLs) in combination with PD-1 inhibitor in treating breast cancer stem-like cells (BCSCs) and drug-resistance breast cancer (DRBC) mice. The results showed that OCT4 and SOX2 were associated with poor prognosis in breast cancer patients. In vitro experiments demonstrated that OCT4&SOX2 CTLs had enhanced cytotoxic activity against BCSCs, which was further improved by the addition of PD-1 inhibitor. In vivo experiments showed that the combination of OCT4&SOX2 CTLs and PD-1 inhibitor reduced tumor volume and weight, and increased tumor apoptosis rate.

HULC targets the IGF1R-PI3K-AKT axis in trans to promote breast cancer metastasis and cisplatin resistance

Summary: Insulin-like growth factor I receptor (IGF1R) is frequently upregulated in breast cancer, and its aberrant activation can enhance tumor cell proliferation, cancer stemness, tumor relapse, metastasis, and chemotherapy resistance. This study identified long non-coding RNA (lncRNA) HULC as a key regulator of IGF1R in breast cancer cells. HULC activates the IGF1R signaling pathway by binding to the intragenic regulatory elements of the gene, promoting intra-chromosomal interactions and histone acetylation. Activation of the HULC-IGF1R/PI3K/AKT pathway leads to tumor resistance to cisplatin through increased expression of cancer stemness markers. In immunodeficient mice, stimulation of the HULC-IGF1R pathway promotes tumor metastasis.

Identification of the ultrahigh-risk subgroup in neuroblastoma cases through DNA methylation analysis and its treatment exploiting cancer metabolism

Summary: This study analyzed DNA methylation and expression datasets of neuroblastomas to identify candidate genes associated with aggressive features. The gene PHGDH, related to serine metabolism, was found to have strong expression and characteristic methylation in a subgroup of neuroblastoma cases. Inhibition of PHGDH suppressed neuroblastoma cell proliferation both in vitro and in vivo, suggesting it as a potential therapeutic alternative. Additionally, inhibiting arginine metabolism showed a combination effect, especially on extracellular arginine-dependent neuroblastoma cells with ASS1 deficiency. The results highlight the rational therapeutic strategy of targeting serine/arginine metabolism for intractable neuroblastoma.

HDAC6 inhibition decreases leukemic stem cell expansion driven by Hedgehog hyperactivation by restoring primary ciliogenesis

Summary: Aberrant activation of the Hh pathway promotes cell proliferation and multi-drug resistance (MDR) in several cancers, including AML. In this study, a positive correlation between Hh, HDAC6, and MDR genes was observed in AML patients, leukemic cell lines, and a zebrafish model. Hyper-activation of Hh or HDAC6 increased proliferation of HSPCs, which could be rescued by HDAC6 inhibition. The presence of a cross-talk between Hh and HDAC6 mediated by stabilization of the primary cilium was detected for the first time. Inhibition of HDAC6 activity alone or in combination with cytarabine efficiently rescued the hematopoietic phenotype, suggesting HDAC6 as a potential therapeutic target for reducing proliferation of leukemic blasts in AML patients.
