Restoring myocardial infarction-induced long-term memory impairment by targeting the cystic fibrosis transmembrane regulator

Summary: Our study investigated the mechanisms that alter hippocampal neurons following myocardial infarction (MI) and explored the therapeutic potential of correcting cystic fibrosis transmembrane regulator (CFTR) as an intervention. We found that MI leads to reduced hippocampal dendrite length and spine density, which is associated with decreased neuronal CFTR expression and inflammatory responses. Blocking CFTR activity down-regulates synaptic regulator PSD-95 expression in neurons, while pharmacologically correcting CFTR expression rescues the down-regulation. Increasing hippocampal neuron CFTR expression improves MI-associated alterations in neuronal structure and memory function. These findings suggest that CFTR therapeutics can attenuate cognitive impairment in heart failure patients.

Summary: This study revealed the significant role of DYRK1A in cardiomyocyte cell cycle activation and cardiac repair. The deletion or inhibition of DYRK1A induced cardiomyocyte cell cycle activation and improved cardiac function. Transcriptomic and epigenomic analyses demonstrated enhanced expression of cell proliferation-related genes and histone modifications in response to DYRK1A depletion. Additionally, two proteins, WD repeat-containing protein 82 and lysine acetyltransferase 6A, were identified as key mediators in the epigenetic modification and pro-proliferative activity of cardiomyocytes.

Summary: This study demonstrated that caffeic acid (CA) can effectively enhance the detection and imaging of high-molecular-weight (HMW) proteins in biological tissue sections through matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Compared to sinapinic acid (SA) and ferulic acid (FA), CA showed superior performance in detecting HMW proteins in biological tissues. Additionally, the use of a CA matrix significantly improved the imaging of proteins on selected tissue sections, demonstrating its potential as a standard organic acid matrix for tissue imaging of HMW proteins.
