Improving cassava bacterial blight resistance by editing the epigenome

Kira M. Veley et al.

Summary: The targeted methylation at the TAL20 effector binding element of the cassava SWEET10a gene leads to resistance to Xanthomonas phaseoli. This study demonstrates that blocking access to host susceptibility (S) genes through targeted methylation can increase disease resistance.

Emerging Roles of SWEET Sugar Transporters in Plant Development and Abiotic Stress Responses

Tinku Gautam et al.

Summary: Sugars are a vital energy source in living organisms and have various functions in osmotic regulation, cell signaling, and energy storage. SWEETs, the newest family of sugar transporters, facilitate the diffusion of sugar molecules across cell membranes. In plants, SWEETs play important roles in processes such as phloem loading, senescence, pollen nutrition, grain filling, and response to abiotic and biotic stress. This review summarizes the functions of SWEET transporters in plant development and abiotic stress, as well as discusses the gene expression dynamics of different SWEET transporters under various abiotic stresses. Additionally, the potential of genome editing tools for improving SWEET genes is explored. Overall, the recent advancements in SWEET research have significant implications for crop improvement and abiotic stress tolerance.

CRISPR/Cas9-Targeted Knockout of Rice Susceptibility Genes OsDjA2 and OsERF104 Reveals Alternative Sources of Resistance to Pyricularia oryzae

Fabiano T. P. K. T. avora et al.

Summary: This study reports that knockout of rice susceptibility genes OsDjA2 and OsERF104 significantly improves blast resistance in rice. The CRISPR/Cas9-mediated gene mutation resulted in stable T0 transformants, and the phenotyping of T1 mutant lines showed a significant improvement in blast resistance compared to the infected control plants.

Consequences of adaptation of TAL effectors on host susceptibility to Xanthomonas

Doron Teper et al.

Summary: By using an experimental evolution approach, we identified adaptive mutations and deletions in TALEs that cannot recognize the EBE of host targets, which reconstituted virulence on the host in less than 30 cycles of repeated infections. Adaptive variants originated from TALEs with a small number of mismatches to the EBE, while TALEs with more mismatches failed to adapt during the study period. This study experimentally demonstrates adaptive rearrangements of TALEs during host adaptation and highlights the importance of considering resistance durability in modified crops introduced into the field.

Mutations introduced in susceptibility genes through CRISPR/Cas9 genome editing confer increased late blight resistance in potatoes

Nam Phuong Kieu et al.

Summary: The use of pathogen-resistant cultivars is expected to increase yield and decrease fungicide use in agriculture. However, in potato breeding, increased resistance obtained via resistance genes (R-genes) is hampered because R-gene(s) are often specific for a pathogen race and can be quickly overcome by the evolution of the pathogen.

A centenary for bacterial spot of tomato and pepper

Ebrahim Osdaghi et al.

Summary: The disease symptoms include water-soaked areas, chlorosis, and necrotic spots on plants, with host range mainly on tomatoes and peppers. The pathogen belongs to Xanthomonas genus and has synonyms like Bacterium vesicatorium, with gram-negative colonies found in general culture media. It is widespread in Africa, Brazil, Canada, the USA, Australia, eastern Europe, and south-east Asia.

Loss of function of a DMR6 ortholog in tomato confers broad-spectrum disease resistance

Daniela Paula de Toledo Thomazella et al.

Summary: The study reveals that disabling susceptibility (S) genes is a promising alternative to confer durable and broad-spectrum disease resistance in breeding programs. In tomato, SlDMR6-1 shows enhanced resistance to various pathogens when up-regulated, accompanied by increased salicylic acid (SA) levels and transcriptional activation of immune responses.
