Regulation of PIEZO1 channels by lipids and the structural components of extracellular matrix/cell cytoskeleton

Valeria Vasileva et al.

Summary: PIEZO1 is a mechanosensitive channel widely found in eukaryotic organisms. The molecular structure, activation mechanisms, and regulating pathways of PIEZO1 remain open questions. Two hypotheses - force-from-lipids and force-from-filaments - have been proposed to explain PIEZO1 gating. Both lipids and components of the extracellular matrix (ECM) and cytoskeleton have been shown to play important roles in the regulation of PIEZO1 properties. The microenvironment of the channel, including the plasma membrane, cytoskeleton, and ECM, is critical for the functioning of PIEZO1.

The plasticity of fibroblasts: A forgotten player in the aging process

Antero Salminen

Summary: Tissue-resident fibroblasts possess plasticity and can modify their properties according to the microenvironment. Different phenotypes of fibroblasts are associated with various tissue pathological conditions, and they can be subdivided into fibrogenic, nonfibrogenic, inflammatory, immunosuppressive, and senescent subsets. The differentiation of myofibroblasts, a major activated fibroblast phenotype, can be induced by multiple stresses associated with aging, and anti-aging treatments can inhibit this differentiation. The versatile plasticity and structural importance of fibroblasts suggest that they play a significant role in the aging process.

Targeting extracellular matrix glycation to attenuate fibroblast activation

Minjeong Jang et al.

Summary: In this study, the researchers found that accumulated AGEs in the ECM can alter the phenotype of fibroblast within a three-dimensional collagen matrix, leading to the acquisition of a cancer-associated fibroblast (CAF)-like phenotype. These effects can be blocked by neutralizing antibodies against the AGE receptor (RAGE) or by inhibiting focal adhesion signaling. In addition, an AGE cross-link breaker called ALT-711 can reduce the transformation of fibroblasts into the CAF-like phenotype. Decreased matrix stiffness also attenuates fibroblast activation. These findings suggest potential therapeutic strategies targeting non-enzymatic ECM glycation to regulate fibroblast activation.

DDR1 associates with TRPV4 in cell-matrix adhesions to enable calcium-regulated myosin activity and collagen compaction

Mechanical stretch promotes hypertrophic scar formation through mechanically activated cation channel Piezo1

Jiahao He et al.

Summary: The study identified that Piezo1 channel is overexpressed in myofibroblasts of hypertrophic scar (HS) tissues and plays a regulatory role in fibroblasts, thus participating in HS formation.
