Intraspecific diversity in the mechanisms underlying abamectin resistance in a cosmopolitan pest

Summary: Pesticide resistance in the cosmopolitan pest Tetranychus urticae involves various mechanisms, including target-site insensitivity, metabolic detoxification, and additive or synergistic effects. In this study, researchers used experimental evolution and bulked segregant analysis to map quantitative trait loci (QTL) associated with abamectin resistance in two populations of T. urticae. They found shared QTLs containing genes encoding subunits of the glutamate-gated chloride channel (GluCl), as well as novel candidate loci. The study also revealed a non-functional variant of GluCl2 as a unique resistance mechanism. This research uncovers the complex basis of abamectin resistance and highlights the diversity of genetic mechanisms underlying resistance in this pest.

Overexpression of PxαE14 Contributing to Detoxification of Multiple Insecticides in Plutella xylostella (L.)

Summary: In this study, a carboxylesterase gene, Px alpha E14, was found to be significantly overexpressed in a field-evolved multiresistant population of the diamondback moth, Plutella xylostella. Px alpha E14 was predominantly expressed in the midgut and malpighian tubule of larvae. Knockdown of Px alpha E14 increased the susceptibility of the larvae to multiple insecticides, while overexpression of Px alpha E14 in fruit flies increased their tolerance to these insecticides. Recombinant Px alpha E14 expressed in Escherichia coli exhibited metabolic activity against multiple insecticides. These findings contribute to our understanding of the molecular mechanisms underlying multiresistance in insect pests.

Pymetrozine-resistant brown planthopper showed cross-resistance with some neonicotinoids such as dinotefuran, nitenpyram, and sulfoxaflor. The resistance to pymetrozine in N. lugens was found to be polygenic, autosomal, and incompletely dominant. The fitness costs associated with pymetrozine resistance can be utilized in insect resistance management strategies to prolong the effectiveness of pymetrozine for controlling N. lugens in China.

Additionally, increased GST activity in abamectin-resistant T. urticae populations appears to be part of a compensatory feedback loop to counteract the effects of the acaricide.

Genome mapping coupled with CRISPR gene editing reveals a P450 gene confers avermectin resistance in the beet armyworm

This study reports a high-quality chromosome-level assembly of the beet armyworm genome and identifies the locus of avermectin resistance using BSA, with further confirmation of the involvement of CYP9A186 gene through gene editing. The natural substitution in the CYP9A186 protein results in enhanced metabolism of avermectins, highlighting the importance of coupling gene editing with BSA for identifying metabolic resistance genes.
