Radical C-H Alkylation with Ethers and Unactivated Cycloalkanes toward the Assembly of Tetrasubstituted Amino Acid Derivatives

Marcos San Segundo et al.

Summary: This study describes a radical alpha-C-H alkylation of a collection of N-picolinamide amino acid derivatives using ethers and cycloalkanes as chemical feedstock. This method is characterized by its reliable scalability and removable auxiliary group, and allows for the assembly of various tri- and tetrasubstituted amino acid compounds.

Modular Synthesis of α,α-Diaryl α-Amino Esters via Bi(V)-Mediated Arylation/SN2-Displacement of Kukhtin-Ramirez Intermediates

Alessio Calcatelli et al.

Summary: We report a concise and modular approach to synthesizing alpha,alpha-diaryl alpha-amino esters from readily available alpha-keto esters. This method proceeds via ketone umpolung and involves the in situ formation of a Kukhtin-Ramirez intermediate, followed by sequential electrophilic arylation by Bi(V) and SN2 displacement by an amine. It is compatible with a wide range of anilines and primary amines, including derivatives of drugs and proteinogenic amino acids, as well as Bi(V) arylating agents and alpha-keto ester substrates.

Phosphine-catalyzed γ-addition of nitroacetates to allenoates for enantioselective creation of α,α-disubstituted α-amino acid precursors

Kaizhi Li et al.

Summary: Enantioselective gamma-addition of readily available alpha-substituted nitroacetates to allenoates has been achieved, providing a straightforward asymmetric synthesis of optically enriched alpha,alpha-disubstituted alpha-amino acid precursors. The reaction proceeds efficiently with high yields and good to excellent enantioselectivities in the presence of amino acid-derived phosphine catalysts, forming products containing a tetra-substituted stereogenic center.
