Gold-catalyzed formal (3+2) and (4+2) cycloadditions of alkynes to highly functionalized dihydropyrroles and tetrahydropyridines

Summary: This research reports a new gold-catalyzed formal cycloaddition reaction that utilizes readily available alkynes and amino but/pent-2-enoate derivatives as reactants to synthesize highly functionalized 2,3-dihydropyrroles and 1,2,3,4-tetrahydropyridines. The reaction shows high efficiency and a broad substrate scope, and can be easily performed at the millimole scale. The resulting products can be further transformed into other valuable N-heterocycles.

Nickel-Catalyzed Chemoselective Arylation of Amino Alcohols

Summary: A systematic evaluation of competitive bisphosphine/Ni-catalyzed C-N versus C-O cross-couplings involving model compounds enabled the development of selective O- and N-arylation of amino alcohols with (hetero)aryl chloride electrophiles, without the need for protection group chemistry. The use of the CyPAd-DalPhos pre-catalyst C2 allowed for challenging O-arylation in amino alcohols with branched alkylamine groups, while selective N-arylation was observed in substrates with less hindered linear alkylamine and aniline groups. The reaction scope in (hetero)aryl chloride was broad, and the transformations could be conducted using benchtop handling of materials.

In this study, a gold(I)-catalyzed heteroannulation of salicylic amides with alkynes was developed, leading to a wide range of 1,3-benzoxazin-4-ones. The utility of this protocol was demonstrated by synthesizing variously substituted benzoxazinones containing quaternary carbon centers, displaying high functional group tolerance and excellent atom economy.
