Impact of Age on Prognosis in Papillary Thyroid Carcinoma: How Should Age be Incorporated into the Treatment Strategy?

Kiminori Sugino et al.

Summary: Age is an important prognostic indicator for thyroid cancer. This study found that age, extrathyroidal extension, and numbers of metastatic lymph nodes are related to disease-specific survival and distant metastasis-free survival in papillary thyroid cancer (PTC) patients. Age does not significantly affect disease-free survival in patients without extrathyroidal extension. However, in patients with extrathyroidal extension, elderly patients have worse survival rates compared to young patients. Therefore, age should be considered when determining the surgical strategy for PTC patients.

Hsa_Circ_0000826 inhibits the proliferation of colorectal cancer by targeting AUF1

Zheying Zhang et al.

Summary: We report a circRNA named hsa_Circ_0000826 (Circ_0000826) that is significantly downregulated in colorectal cancer (CRC) tissues and is associated with a poor prognosis in patients. Silencing Circ_0000826 promotes CRC cell proliferation, while overexpression of Circ_0000826 inhibits CRC cell proliferation. Furthermore, Circ_0000826 can bind to AUF1 and competitively inhibit its binding to c-MYC 30-UTR, leading to decreased c-MYC expression and cell proliferation.

Thyroid Cancer Incidence and Mortality Trends in the United States: 2000-2018

Uchechukwu C. C. Megwalu et al.

Summary: Thyroid cancer incidence rates have declined since 2014, with similar declines noted for papillary thyroid carcinoma (PTC), localized disease, and tumors <= 1.0 cm, but not for tumors >2.0 cm. Incidence-based mortality rates continue to increase. These findings suggest changes in the management of thyroid nodules may have led to a decrease in the diagnosis of small indolent tumors, but not more advanced tumors.

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

CircRNA circTIAM1 promotes papillary thyroid cancer progression through the miR-646/HNRNPA1 signaling pathway

Deguang Zhang et al.

Summary: In this study, circRNA high-throughput sequencing was used to identify alterations in circRNA expression levels in PTC tissues. circTIAM1 was found to be upregulated in PTC and associated with apoptosis, proliferation, and migration of PTC cells. Mechanistically, circTIAM1 acted as a sponge of microRNA-646 and functioned in PTC by targeting miR-646 and heterogeneous ribonucleoprotein A1. These findings highlight the oncogenic role of circTIAM1 in PTC and suggest its potential as a therapeutic target for PTC progression.

Tumor-derived exosomal circPSMA1 facilitates the tumorigenesis, metastasis, and migration in triple-negative breast cancer (TNBC) through miR-637/Akt1/β-catenin (cyclin D1) axis

Su-jin Yang et al.

Summary: This study reveals that circPSMA1 functions as a tumor promoter in triple-negative breast cancer (TNBC) by acting as a miRNA sponge to absorb miR-637 and regulate Akt1-beta-catenin (cyclin D1) axis. Exosomes from TNBC cells carrying high levels of circPSMA1 can transmit migration and proliferation capacity to recipient cells. High expression of Akt1 and low expression of miR-637 are highly correlated with poor prognosis in TNBC patients with lymph node metastasis.
