Summary: Age is an important prognostic indicator for thyroid cancer. This study found that age, extrathyroidal extension, and numbers of metastatic lymph nodes are related to disease-specific survival and distant metastasis-free survival in papillary thyroid cancer (PTC) patients. Age does not significantly affect disease-free survival in patients without extrathyroidal extension. However, in patients with extrathyroidal extension, elderly patients have worse survival rates compared to young patients. Therefore, age should be considered when determining the surgical strategy for PTC patients.

Summary: We report a circRNA named hsa_Circ_0000826 (Circ_0000826) that is significantly downregulated in colorectal cancer (CRC) tissues and is associated with a poor prognosis in patients. Silencing Circ_0000826 promotes CRC cell proliferation, while overexpression of Circ_0000826 inhibits CRC cell proliferation. Furthermore, Circ_0000826 can bind to AUF1 and competitively inhibit its binding to c-MYC 30-UTR, leading to decreased c-MYC expression and cell proliferation.

Summary: This study identified circNEIL3 (hsa_circ_0001460) as a TGF beta-repressive and metastasis-related circRNA, which effectively inhibits tumor metastasis by directly interacting with onco-genic protein YBX1 and promoting its Nedd4L-mediated proteasomal degradation. The findings highlight the significance of circNEIL3 as a potential biomarker and therapeutic option in cancer treatment, and shed light on the molecular mechanism of circRNA in metastasis.

Summary: This study identifies the role of the purine biosynthesis pathway enzyme ATIC in regulating LRRK2 protein levels and toxicity. The precursor AICAr can regulate LRRK2 levels in a cell-type-specific manner in vitro and in mouse tissue. AICAr regulates LRRK2 levels through AUF1-mediated mRNA decay. This study provides insights into a novel regulatory mechanism of LRRK2 protein levels and function, independent of its enzymatic activity.

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

Summary: Thyroid cancer incidence rates have declined since 2014, with similar declines noted for papillary thyroid carcinoma (PTC), localized disease, and tumors <= 1.0 cm, but not for tumors >2.0 cm. Incidence-based mortality rates continue to increase. These findings suggest changes in the management of thyroid nodules may have led to a decrease in the diagnosis of small indolent tumors, but not more advanced tumors.

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Summary: In this study, circRNA high-throughput sequencing was used to identify alterations in circRNA expression levels in PTC tissues. circTIAM1 was found to be upregulated in PTC and associated with apoptosis, proliferation, and migration of PTC cells. Mechanistically, circTIAM1 acted as a sponge of microRNA-646 and functioned in PTC by targeting miR-646 and heterogeneous ribonucleoprotein A1. These findings highlight the oncogenic role of circTIAM1 in PTC and suggest its potential as a therapeutic target for PTC progression.

CircARID1A was significantly upregulated in gastric cancer tissues and positively correlated with tumor characteristics. Knocking down circARID1A inhibited the proliferation of gastric cancer cells. Mechanistically, circARID1A acted as a scaffold to facilitate the interaction between IGF2BP3 and SLC7A5 mRNA, thereby increasing the stability of SLC7A5 mRNA and promoting gastric cancer proliferation through the AKT/mTOR pathway.

Summary: This study found that high expression of circ_0011385 in HCC is closely associated with tumor size, TNM stage, and prognosis, and its inhibition can prevent proliferation of HCC cells. Circ_0011385 acts as a sponge for miR-361-3p, regulating the mRNA expression of STC2, with its transcription being regulated by SP3. Therefore, circ_0011385 may serve as a new biomarker for HCC.

Summary: This study reveals that circPSMA1 functions as a tumor promoter in triple-negative breast cancer (TNBC) by acting as a miRNA sponge to absorb miR-637 and regulate Akt1-beta-catenin (cyclin D1) axis. Exosomes from TNBC cells carrying high levels of circPSMA1 can transmit migration and proliferation capacity to recipient cells. High expression of Akt1 and low expression of miR-637 are highly correlated with poor prognosis in TNBC patients with lymph node metastasis.
