LYL1 facilitates AETFC assembly and gene activation by recruiting CARM1 in t(8;21) AML

Qian Chen et al.

Summary: The AML1-ETO fusion protein is a key factor in acute myeloid leukemia, with its activity dependent on the LYL1 component in the AETFC complex. Through interaction with CARM1, LYL1 facilitates the binding of AETFC to active enhancers, leading to the activation of AML1-ETO-dependent gene expression.

WhichTF is functionally important in your open chromatin data?

Yosuke Tanigawa et al.

Summary: WhichTF is a computational method that can identify functionally important transcription factors (TFs) from chromatin accessibility measurements. It integrates accessibility measurements, high-confidence pre-computed conservation-aware TF binding sites, and putative gene-regulatory models to rank TFs. By comparing with other methods, WhichTF demonstrates its unique ability to identify context-specific TFs with functional relevance. It also applies differential analysis to distinguish the transcriptional regulatory landscape in closely related samples and identifies potential novel TFs involved in various biological processes and diseases. WhichTF offers insights into TF-mediated transcriptional regulation in different contexts.

BCOR and BCORL1 Mutations Drive Epigenetic Reprogramming and Oncogenic Signaling by Unlinking PRC1.1 from Target Genes

Eva J. Schaefer et al.

Summary: We demonstrate that mutations of PRC1 subunits BCOR and BCORL1 in leukemia lead to epigenetic reprogramming and activation of aberrant cell signaling programs, resulting in resistance to treatment. Our findings provide mechanistic insights into the pathogenesis of PRC1.1-mutated leukemia and suggest potential therapeutic approaches.

RUNX1/RUNX1T1 mediates alternative splicing and reorganises the transcriptional landscape in leukemia

Vasily V. Grinev et al.

Summary: The fusion oncogene RUNX1/RUNX1T1 plays a key role in acute myeloid leukemia by regulating alternative RNA splicing in leukemic cells. This regulation is achieved through changing 5'UTR structure, altering isoform expression, and controlling the expression of splicing factors. This study highlights the importance of alternative splicing in leukemia transcriptome re-organization by an aberrant transcriptional regulator.

The leukemic oncogene EVI1 hijacks a MYC super-enhancer by CTCF-facilitated loops

Sophie Ottema et al.

Summary: Chromosome rearrangements can result in altered oncogene expression in cancer, such as the overexpression of EVI1 due to a 3q26 translocation. The 'enhancer hijacking' mechanism facilitated by CTCF-mediated loops is found to be responsible for this overexpression. This process is observed in therapy-resistant AML cases and involves the translocation of a MYC super-enhancer to the EVI1 locus.

Core-binding factor leukemia hijacks the T-cell-prone PU.1 antisense promoter

E. van der Kouwe et al.

Summary: The study reveals that downregulation of PU.1 in the blood system is an active process induced by an antisense promoter driven by RUNX transcription factors, resulting in myeloid differentiation blockade. Patients with CBF-AML show an elevated antisense/sense transcript ratio and promoter accessibility compared to normal karyotype AML or healthy CD34(+) cells. Additionally, competitive interaction between an enhancer and the proximal or antisense promoter acts as a binary switch for myeloid or T-cell development.

Disruption of a GATA2-TAL1-ERG regulatory circuit promotes erythroid transition in healthy and leukemic stem cells

Julie A. I. Thoms et al.

Summary: This study reveals the presence of the fully connected heptad circuit in both healthy hematopoietic stem and progenitor cells (HSPCs) and leukemic cells, with only minor alterations in acute myeloid leukemia (AML). The subcircuit consisting of GATA2, TALI, and ERG regulates stem cell-to-erythroid transition in both healthy and leukemic cells, suggesting the potential for manipulating these factors to facilitate specific cell-type transitions and overcome dysregulated hematopoiesis.

Enhancer Hijacking Drives Oncogenic BCL11B Expression in Lineage-Ambiguous Stem Cell Leukemia

Lindsey E. Montefiori et al.

Summary: This study identified a subgroup of acute leukemias with ambiguous lineage expressing myeloid, T lymphoid, and stem cell markers, driven by aberrant allele-specific deregulation of the master transcription factor BCL11B. Chromosomal rearrangements and focal amplifications leading to superenhancer generation were identified as mechanisms behind the oncogenic deregulation of BCL11B. This ectopic expression of BCL11B in hematopoietic cells mediated by enhancer hijacking was suggested as an oncogenic driver of human lineage-ambiguous leukemia.
