Summary: Super-resolution imaging reveals that RAD21 plays a unique role in cohesin loading and chromatin structure regulation. Up-regulation of RAD21 leads to chromatin extrusion into a vermicelli-like morphology and beads-on-a-string-type pattern, while up-regulation of other cohesin subunits results in even distributions. Mechanistically, RAD21 facilitates cohesin loading through RAD21-loader interaction, and its aberrant high expression is associated with poor patient survival in breast cancer.

Mapping genomic loci implicates genes and synaptic biology in schizophrenia

Polycomb-mediated genome architecture enables long-range spreading of H3K27 methylation

Summary: Polycomb-group proteins are involved in gene silencing through H3K27me3 deposition and chromatin compaction. This study investigates the distribution of long-range Polycomb-associated DNA loops in mouse ESCs and human induced pluripotent stem cells, and finds that H3K27me3 loop anchors are enriched for Polycomb nucleation points and coincide with key developmental genes. Furthermore, the study demonstrates that RNA binding is crucial for PRC2 occupancy and Polycomb-associated DNA looping. These findings reveal the importance of developmental gene loci in Polycomb spreading and provide insights into the regulation of gene silencing.

Cohesin-dependence of neuronal gene expression relates to chromatin loop length

Summary: Cohesin plays a significant role in the expression of genes involved in neuronal maturation and homeostasis. Different classes of activity-regulated genes in mouse primary cortical neurons have distinct reliance on cohesin. Cohesin is required for full expression of certain secondary response genes with long-range chromatin contacts and also scales with the genomic distance traversed by chromatin loops.

LncRNA RUS shapes the gene expression program towards neurogenesis

Summary: The evolution of brain complexity is correlated with the increased expression of long noncoding RNAs in neural tissues. In this study, the functional characterization of the lncRNA LINC01322, also known as RUS, was presented. RUS is exclusively expressed in neural cells and plays a role in neuronal differentiation. Depletion of RUS results in arrest of neuronal precursors and changes in the expression of genes involved in neurogenesis. This suggests that RUS may mediate gene activation and repression in a context-dependent manner.

Enhancer release and retargeting activates disease-susceptibility genes

This process of target-switching may be determined by CTCF binding at promoters, in line with the model of 'enhancer scanning'.
