Avoiding misleading estimates using mtDNA heteroplasmy statistics to study bottleneck size and selection

Konstantinos Giannakis et al.

Summary: Mitochondrial DNA heteroplasmy samples are essential for understanding the developmental and genetic processes of mitochondrial DNA populations. However, using sample statistics and variance can lead to misleading results in hypothesis testing and bottleneck size estimates. We propose a maximum likelihood approach that addresses these issues and emphasizes the use of explicit hypothesis tests.

Altered collective mitochondrial dynamics in the Arabidopsis msh1 mutant compromising organelle DNA maintenance

Joanna M. Chustecki et al.

Summary: This study shows that plant mitochondria move to facilitate potential exchange between organelles, and that this movement is influenced by genetic and physical factors. The altered mitochondrial dynamics observed in the msh1 mutant suggest a compensatory mechanism for maintaining genetic structure.

Evolutionary inference across eukaryotes identifies universal features shaping organelle gene retention

Konstantinos Giannakis et al.

Summary: This study utilized over 15,000 oDNA sequences and over 300 whole genome sequences across eukaryotes to identify the properties of genes and mechanisms that determine oDNA retention. The findings suggest that factors such as protein hydrophobicity and binding energy centrality within a protein complex play important roles in oDNA retention.

Phylodynamics for cell biologists

T. Stadler et al.

Summary: Multicellular organisms are made up of cells connected through ancestry. The dynamics of cell processes in an organism result in development, differentiation, and cancer. Advances in molecular biology now allow for the study of individual cells within an organism, with theoretical and experimental progress needed to fully utilize the information in single-cell data.

Avoiding organelle mutational meltdown across eukaryotes with or without a germline bottleneck

David M. Edwards et al.

Summary: A comprehensive genetic model has been constructed to explain how different mechanisms segregate and decrease damage in organelle DNA (oDNA) across eukaryotes. Gene conversion is identified as a powerful mechanism to increase beneficial cell-to-cell variance without depleting oDNA copy number, explaining the advantage of observed oDNA recombination in diverse organisms. This framework provides insights into how eukaryotes avoid Muller's ratchet and explains the puzzling oDNA differences across taxa.

Network analysis of Arabidopsis mitochondrial dynamics reveals a resolved tradeoff between physical distribution and social connectivity

Joanna M. Chustecki et al.

Summary: This study reveals the tradeoff between physical priorities and "social" priorities of mitochondria dynamics in Arabidopsis, as well as how plant cells resolve this tension to favor efficient networks with high potential for exchanging contents. The combination of physical modeling and experimental data through network analysis sheds light on the fundamental principles underlying complex organelle dynamics.

Nuclear-cytoplasmic balance: whole genome duplications induce elevated organellar genome copy number

Matheus Fernandes Gyorfy et al.

Summary: The study reveals that in plant whole genome duplication events, the imbalance in copy numbers between nuclear and organellar genomes can trigger an immediate cellular response, with polyploids maintaining an elevated organellar genome copy number trend despite changes in nuclear genome copy number. Maintenance of cytonuclear stoichiometry is identified as a crucial component in the establishment of polyploid lineages.
