Rehabilitation of a historically contaminated soil by different laccases and laccase-mediator system

Ziva Vipotnik et al.

Summary: This study evaluated the degradation of polyaromatic hydrocarbons (PAH) in naturally contaminated soil using laccases. The results showed that laccases, with or without mediators, can effectively degrade PAH contaminants in soil, providing a potential approach for soil remediation.

Surfing the wave of oxyfunctionalization chemistry by engineering fungal unspecific peroxygenases

Alejandro Beltran-Nogal et al.

Summary: The selective insertion of oxygen into non-activated organic molecules is crucial for the synthesis of existing and next generation industrial chemicals or pharmaceuticals. Fungal unspecific peroxygenases (UPOs) are highly active biocatalysts for C-H oxyfunctionalization, but their limited availability has hindered their use in industry. However, the combination of directed evolution and computational design is bringing UPOs closer to practical applications.

Ligninolytic enzymes production during polycyclic aromatic hydrocarbons degradation: effect of soil pH, soil amendments and fungal co-cultivation

Ziva Vipotnik et al.

Summary: Soil microorganisms play a crucial role in the degradation of PAHs through various metabolic pathways. The study evaluated the impact of soil pH, soil amendments, and fungal co-cultivation on PAH degradation and ligninolytic enzyme activity. Results showed that fungal remediation could effectively degrade 85-90% of PAHs, and co-cultivation could enhance the degradation rates of certain PAHs, although higher enzyme production was observed when fungi were cultivated alone.

Pilot-Scale Production of Peroxygenase from Agrocybe aegerita

Fabio Tonin et al.

Summary: The pilot-scale production of the peroxygenase from Agrocybe aegerita (rAaeUPO) is demonstrated, resulting in a final concentration of 0.29 g L-1 after 6 days of fermentation. However, only 170 g of the enzyme has been isolated due to nonoptimized downstream processing. The enzyme preparation showed preparative usefulness at a semipreparative scale for the stereoselective hydroxylation of ethyl benzene, achieving more than 340,000 catalytic turnovers and producing over 5 g L-1 (R)-1-phenyl ethanol in 7 days.
