A sensory neuron-specific long non-coding RNA reduces neuropathic pain by rescuing KCNN1 expression

Bing Wang et al.

Summary: This study identifies a sensory neuron-specific long non-coding RNA, SS-lncRNA, which is predominantly expressed in small neurons of the dorsal root ganglion and contributes to the development and maintenance of neuropathic pain by regulating KCNN1 expression. The downregulation of SS-lncRNA leads to a decrease in KCNN1 expression and the development of neuropathic pain symptoms.

CBX2 shapes chromatin accessibility promoting AML via p38 MAPK signaling pathway

Nunzio Del Gaudio et al.

Summary: CBX2 plays a crucial role in leukemia progression, and its silencing can inhibit cell proliferation and induce apoptosis, as well as alter cell transcriptional programs. CBX2 suppression also increases chromatin accessibility, alters gene expression, and results in enrichment of cell death pathways and downregulation of survival genes. CBX2 silencing also induces epigenetic reprogramming at p38 MAPK-associated regulatory sites, leading to dysregulation of gene expression.

ZNF382 controls mouse neuropathic pain via silencer-based epigenetic inhibition of Cxcl13 in DRG neurons

Longfei Ma et al.

Summary: Injury-induced down-regulation of ZNF382 in DRG neurons causes neuropathic pain, which can be alleviated by CXCL13 silencing or CXCR5 knockout. ZNF382 down-regulation suppresses Cxcl13 transcription through a cis-acting silencer, and disrupting this silencer leads to Cxcl13 transcriptional activation and neuropathic pain. This down-regulation of ZNF382 may be essential for the development of neuropathic pain through epigenetic disinhibition of CXCL13 in DRG neurons.

Methyltransferase-like 3 contributes to inflammatory pain by targeting TET1 in YTHDF2-dependent manner

Zhiqiang Pan et al.

Summary: The study reveals that downregulation of spinal METTL3 and coordination with YTHDF2 contribute to the modulation of inflammatory pain through stabilizing upregulation of TET1 in spinal neurons. Manipulating Mett13 levels can affect pain behavior, and decreased levels of YTHDF2 can lead to increased TET1 levels in the spinal cord and influence pain regulation.
