The role of body fat in multiple sclerosis susceptibility and severity: A Mendelian randomisation study

Summary: This study aimed to explore the potential causal associations of anthropometric measures with multiple sclerosis (MS) risk and severity. The results showed that features linked to obesity were risk factors for MS development and severity, while height and non-fat mass were not associated with MS.

Summary: The study found that childhood and adulthood body mass index were positively associated with MS, while physical activity and serum 25-hydroxyvitamin D were inversely associated with MS. Additionally, possible associations were found for type 2 diabetes, waist circumference, body fat percentage, age of puberty, and high-density lipoprotein cholesterol in relation to MS. Data from systematic review suggested that exposure to organic solvents, Epstein Barr virus and cytomegalovirus infection, and diphtheria and tetanus vaccination were also associated with MS risk.

Most genes show cis-eQTL effects, while distal trans-eQTL effects have lower replication rates. However, single-cell RNA-seq data prioritize intracellular trans-eQTL effects mediated by transcription factors. Additionally, a subset of genes correlated with polygenic scores for various phenotypes may serve as potential drivers for those traits.

Summary: The study identified RAC1 as a potential therapeutic target for HCC and demonstrated the potent antitumor effect of the RAC1 inhibitor 1D-142 in highly proliferative HCC. The inhibition of RAC1 by 1D-142 resulted in apoptosis and cell cycle arrest in HCC cells, as well as a decrease in tumor growth and metastasis in HCC mouse models. Additionally, 1D-142 showed an anti-fibrotic effect by decreasing hepatic stellate cell activation in vivo.
