Proton magnetic resonance spectroscopy in frontotemporal lobar degeneration-related syndromes

Alexander G. Murley et al.

Summary: There is an urgent need to better understand the pathophysiology of cognitive impairment in syndromes associated with frontotemporal lobar degeneration. This study used magnetic resonance spectroscopy to quantify metabolite deficits in patients with different clinical syndromes. The findings revealed significant differences in metabolite concentrations among brain regions and diagnoses, and reduced concentrations of N-acetyl aspartate and glutamate were associated with executive dysfunction and behavioral impairment.

Dynamic causal modelling of COVID-19 and its mitigations

Karl J. Friston et al.

Summary: This technical report presents a dynamic causal model for mitigated epidemiological outcomes during the COVID-9 coronavirus outbreak in 2020. It utilizes variational Bayesian procedures to estimate parameters and embeds conventional models within a larger sociobehavioural response model, allowing for assumption-free forecasting.

In Vivo Assay of Cortical Microcircuitry in Frontotemporal Dementia: A Platform for Experimental Medicine Studies

Alexander D. Shaw et al.

Summary: The study focuses on using dynamic causal modeling to transfer canonical microcircuit models into noninvasive human magnetoencephalography for investigating regional- and laminar-specific changes in frontotemporal dementia. The model-based approach demonstrates accurate differentiation between patients and healthy controls, providing a potential platform for testing disease progression hypotheses and pharmacotherapeutics.

GABAergic cortical network physiology in frontotemporal lobar degeneration

Natalie E. Adams et al.

Summary: The study highlighted deficits in GABA levels and physiological abnormalities in patients with frontotemporal lobar degeneration, showing a correlation between GABA impairment and cognitive/behavioral changes. The use of dynamic causal modelling provided insight into mechanisms of human neurodegenerative diseases and variations in response to therapies, suggesting the potential for targeted neurophysiological restoration in selected patients.

Conductance-based dynamic causal modeling: A mathematical review of its application to cross-power spectral densities

Ines Pereira et al.

Summary: Dynamic Causal Modeling (DCM) is a Bayesian framework used to infer hidden neuronal states based on brain activity measurements. While promising for understanding human brain dynamics, DCM variants can be challenging to fully understand due to their complexity and reliance on concepts from multiple fields. Solid theoretical knowledge of the models is crucial to avoid pitfalls in their application and interpretation of results.

Computational models link cellular mechanisms of neuromodulation to large-scale neural dynamics

James M. Shine et al.

Summary: This article demonstrates how biophysical models of large-scale neural dynamics can bridge the gap between neuromodulation at the cellular scale and whole-brain systems dynamics, through advancements in mathematics and related fields. While neuromodulation involves altering the biophysical properties of neurons to support adaptive cognitive functions, changes in cognitive function are not directly caused by individual neuron modulation, but are mediated by population dynamics in neural ensembles. Understanding this multiscale mapping is a challenging but crucial task.

No balance between glutamate plus glutamine and GABA plus in visual or motor cortices of the human brain: A magnetic resonance spectroscopy study

Reuben Rideaux

Summary: Based on MRS studies, there is no significant positive correlation between GABA+ and Glx in the visual and motor cortices, indicating a lack of balance between excitatory and inhibitory neurotransmitters across the brain.
