Summary: This study demonstrates the distinct and redundant biological functions among Pol II elongation factor paralogs through the use of orthogonal acute depletion/inhibition strategies, highlighting the roles of BRD4 and SEC families in Pol II release and pausing, as well as the stronger attenuation of the heat shock response through simultaneous depletion of AFF1 and AFF4.

Structural basis of Integrator-mediated transcription regulation

Isaac Fianu et al.

Summary: Integrator and PP2A collaboratively form a complex that dephosphorylates paused Pol II and cleaves nascent RNA to facilitate transcription termination. Integrator binds Pol II and pausing factors to exclude elongation factors, and positions PP2A to counteract Pol II phosphorylation. Additionally, Integrator endonuclease cleaves RNA near the Pol II active site, enabling DNA release and termination of transcription.

A role for the Cockayne Syndrome B (CSB)-Elongin ubiquitin ligase complex in signal-dependent RNA polymerase II transcription

Juston C. Weems et al.

Summary: The CSB-Elongin ubiquitin ligase pathway not only plays a role in the response to DNA damage, but also has a significant impact on the activation of RNAPII-mediated transcription in various signaling pathways, such as endoplasmic reticulum stress, amino acid starvation, retinoic acid, glucocorticoids, and doxycycline. The pathway is involved in the rapid recruitment of CSB and the Elongin ubiquitin ligase to target genes during transcription, suggesting a direct role in gene regulation. Cells lacking the Elongin subunit Elongin A exhibit delays in RNAPII accumulation and dismissal from target genes following hormone treatments, indicating the importance of this pathway in transcription regulation.

Allosteric transcription stimulation by RNA polymerase II super elongation complex

Ying Chen et al.

Summary: This study presents the cryo-EM structure of ELL2-EAF1 bound to a RNA Pol II elongation complex, showing how SEC delivers P-TEFb to RNA Pol II and stimulates RNA elongation. Functional analysis indicates that the stimulation of RNA elongation requires the dimerization module and the ELL2 linker.

Elongin A regulates transcription in vivo through enhanced RNA polymerase processivity

Yating Wang et al.

Summary: Elongin A is a factor associated with RNAPII which localizes to actively transcribed regions and enhancers, with recruitment levels correlating with transcription levels. Loss of Elongin A may lead to reduced serine2-phosphorylated RNAPII and increased promoter pausing, without significant impact on global transcription or elongation rates. This study provides the first comprehensive analysis of Elongin A's role in transcription regulation in vivo.

Elongin A associates with actively transcribed genes and modulates enhancer RNA levels with limited impact on transcription elongation rate in vivo

M. Behfar Ardehali et al.

Summary: EloA localizes to Pol II transcribed genes and active enhancers in mouse embryonic stem cells, regulating Pol II transcription levels through enhancer RNA synthesis and interacting with the machinery for ribosomal RNA production/processing in the nucleolus. It forms phase-separated condensates and truncation mutations in its intrinsically disordered regions interfere with its nucleolar localization.
