A Threshold-Based Bioluminescence Detector With a CMOS-Integrated Photodiode Array in 65 nm for a Multi-Diagnostic Ingestible Capsule

Qijun Liu et al.

Summary: This article presents a highly miniaturized ingestible electronic capsule for biochemical detection using genetically engineered biosensor bacteria. The integrated circuit (IC) inside the capsule achieves low-power bioluminescence detection and successfully detects an intestinal inflammation biomarker in vitro.

A programmable encapsulation system improves delivery of therapeutic bacteria in mice

Tetsuhiro Harimoto et al.

Summary: This study developed a genetically encoded microbial encapsulation system with tunable and dynamic expression of surface capsular polysaccharides to enhance systemic delivery. The system allowed bacteria to temporarily evade immune attack and be effectively cleared in vivo. The results showed that this strategy increased the maximum tolerated dose of bacteria and improved anti-tumor efficacy.

GUT PHYSIOLOGY The microbiome and gut homeostasis

Jee-Yon Lee et al.

Summary: Changes in gut microbiota composition are linked to various human diseases. However, defining homeostasis or dysbiosis based on the presence or absence of specific microbial species has been unsuccessful. The adult gut microbiota is regulated by diet and host factors, with oxygen and nitrate availability shaping the bacterial community in different parts of the gut. Poor diet weakens host control mechanisms, potentially leading to dysbiosis. Therefore, quantifying host parameters that control microbial growth could offer alternative approaches to define and remediate dysbiosis.

Autonomous push button-controlled rapid insulin release from a piezoelectrically activated subcutaneous cell implant

Haijie Zhao et al.

Summary: The study presents a new approach for controlling self-sufficient cellular implants through finger pressure on the skin. Pressing the button generates enough energy to release biopharmaceuticals, and the release can be controlled by adjusting the frequency and duration of finger pressure. The study demonstrates the restoration of normal blood glucose levels in a mouse model of type 1 diabetes using this method.

Debaryomyces is enriched in Crohn's disease intestinal tissue and impairs healing in mice

Umang Jain et al.

Summary: The study revealed that Debaryomyces hansenii fungus inhabits incompletely healed intestinal wounds of mice and inflamed mucosal tissues of CD patients, impairing colonic healing. Mechanistically, D. hansenii affects mucosal healing through the myeloid cell-specific type 1 interferon-CCL5 axis.

Magnetic Living Hydrogels for Intestinal Localization, Retention, and Diagnosis

Xinyue Liu et al.

Summary: Research has shown that by developing an ingestible magnetic hydrogel carrier, synthetic microbes can be localized and safely extended in the gastrointestinal tract for health monitoring and sustained drug release. The carrier allows living bacteria to be retained in the intestine for up to seven days, and by attaching a magnet to the abdominal skin, it resists peristaltic waves in the intestine, achieving device retention.
