Long-Duration Response to Levodopa, Motor Learning, and Neuroplasticity in Early Parkinson's Disease

Giorgia Sciacca et al.

Summary: The study aimed to investigate the effects of long-duration response (LDR) on neuroplasticity parameters in Parkinson's disease patients. The results showed that patients with sustained LDR exhibited significant improvements in neuroplasticity parameters after motor learning, while those without LDR did not show any changes. The conclusion is that in early Parkinson's disease patients, sustained LDR may synergistically facilitate adaptive changes in neuroplasticity in basal ganglia and cortical networks with motor learning.

Identification of Parkinson's Disease Subtypes from Resting-State Electroencephalography

Sahar Yassine et al.

Summary: In this study, different sub-phenotypes of Parkinson's disease (PD) were identified based on electrophysiological profiles obtained from resting-state electroencephalography (RS-EEG). These sub-phenotypes showed distinct disruptions in various brain networks and were predictive of disease outcome. EEG features were also able to predict cognitive evolution of PD patients. The identification of these novel PD subtypes based on electrical brain activity signatures has important clinical implications and can support the development of brain-based therapeutic strategies.

Ivan Mysin et al.

Summary: Brain rhythms, particularly theta and gamma coherence, play a crucial role in information processing and cognitive tasks in the neuronal networks of the brain. This article reviews possible mechanisms of coherence and its significance in the hippocampal formation, highlighting the different functions and generation mechanisms of theta and gamma coherence. It emphasizes the importance of synchronized neuronal firing and the transfer of information between different regions of the brain for cooperative information processing.

Altered Cholinergic Innervation in De Novo Parkinson's Disease with and without Cognitive Impairment

Sygrid van der Zee et al.

Summary: This study assessed the cholinergic innervation status in early-stage Parkinson's disease (PD) patients and found that cholinergic innervation changes were associated with cognitive impairment. Regardless of cognitive status, patients showed cholinergic denervation in the posterior cortical regions. Cognitively intact patients exhibited higher cholinergic activity in the cerebellar, frontal, and subcortical regions, suggesting compensatory cholinergic upregulation in early-stage PD. Limited or failing cholinergic upregulation may play an important role in early cognitive impairment in PD.

Sahar Yassine et al.

Summary: Tracking the longitudinal functional brain dysconnectivity in PD patients over 5 years revealed a gradual impairment in alpha 2 and beta frequency bands, strongly correlated with global cognitive decline. Additionally, these disrupted networks were associated with the lateralization of motor symptoms, highlighting potential biomarkers for PD progression.

Summary: In early stages of Parkinson's disease, alterations in EEG functional connectivity were observed, characterized by reduced connectivity in α-β frequency bands, increased connectivity in the γ band, and differences in assortativity coefficient. Network measures analysis helps to understand the complexity of Parkinson's disease physiopathology.

Reduced Resting-State Cortical Alpha Connectivity Reflects Distinct Functional Brain Dysconnectivity in Alzheimer's Disease and Mild Cognitive Impairment

John Preetham Gurja et al.

Summary: This study found distinct abnormal activity patterns in resting state intrinsic functional brain networks in patients with Alzheimer's disease (AD) and mild cognitive impairment (MCI) using EEG analysis. Reduced connectivity and current source density were observed in the patient groups compared to controls, and a positive correlation between connectivity values in specific brain areas and cognitive decline was found only in the AD group.

STN-DBS Induces Acute Changes in β-Band Cortical Functional Connectivity in Patients with Parkinson's Disease

Matteo Conti et al.

Summary: In this study, the short-term effects of subthalamic nucleus deep-brain stimulation (STN-DBS) on beta-band cortical functional connectivity (FC) were investigated in Parkinson's disease (PD) patients. The results showed that PD patients exhibited widespread cortical hyperconnectivity in beta bands, and STN-DBS could acutely reduce beta FC primarily in cortical regions involved in basal ganglia-cortical circuits.

Summary: This study investigated alterations in brain networks in patients with Alzheimer's disease and mild cognitive impairment using a population-specific brain template. Significant changes in graph metrics were found in the frontal gyrus and cerebellum of AD and MCI groups compared to healthy controls, and some of these metrics were correlated with cognitive function and daily activities. Patients with MCI showed unique graph metric alterations in the cerebellum, suggesting potential brain plasticity differences in this group.

Summary: The study found that right fronto-temporal theta synchronization is positively associated with music-evoked pleasantness when listening to unfamiliar music, while inter-hemispheric temporo-parietal theta synchronization is positively associated with pleasantness when listening to familiar music. These results shed light on the possible oscillatory mechanisms underlying fronto-temporal and temporo-parietal connectivity in relation to music-evoked pleasantness and familiarity.

Prodromal Parkinson disease subtypes - key to understanding heterogeneity

Daniela Berg et al.

Summary: Parkinson's disease begins its pathological processes and neurodegeneration long before clinical diagnosis, allowing for identification of individuals likely to develop the disease through risk and prodromal markers. Heterogeneity in clinical and prodromal PD suggests that subtypes exist, differentiated by clinical manifestations, pathomechanisms, and progression patterns. Understanding this variability is crucial for early and accurate diagnosis, as well as for targeting neuroprotective interventions effectively.
