Synthesis of Unprecedented α/β-Alternate (1→4)-Glucans via Stereoselective Iterative Glycosylation

Fuzhu Yang et al.

Summary: Glucans, such as amylose and cellulose, play crucial roles in energy storage and structural function. In this study, a glycosylation protocol was developed to selectively construct 1,2-cis-alpha- and 1,2-trans-beta-glucosidic linkages, leading to the synthesis of unprecedented alpha/beta-alternate (1 -> 4)-glucans. The synthetic amycellulose exhibited an extended ribbon-like conformation, similar to cellulose.

Directed SN2 Glycosylation Employing an Amide-Functionalized 1-Naphthoate Platform Featuring a Selectivity-Safeguarding Mechanism

Xu Ma et al.

Summary: This work introduces a novel catalytic S(N)2 glycosylation method using an amide-functionalized 1-naphthoate platform as a latent glycosyl leaving group. The amide group facilitates the S(N)2 process by directing the attack of the glycosyl acceptor via H-bonding interaction, resulting in stereoinversion at the anomeric center. Additionally, the amide group enables a safeguarding mechanism by trapping oxocarbenium intermediates, minimizing stereorandom S(N)1 processes. This strategy allows for high-yielding synthesis of a wide range of glycosides with stereoinversion, including challenging 1,2-cis-linkage-rich oligosaccharides.

Summary: In this study, it was found that 1,4-alpha-Glucan branching enzyme (GBE) from Geobacillus thermoglucosidans could be highly expressed in food grade strain Bacillus subtilis, and the secretion efficiency was improved by truncating the loop structure at the N-terminus and replacing hydrophobic residues. These findings provide a new perspective for improving protein expression in B. subtilis.

Effect of highly branched α-glucans synthesized by dual glycosyltransferases on the glucose release rate

Moon-Gi Hong et al.

Summary: Increasing alpha-1,6 linkages in starch molecules leads to the synthesis of highly branched alpha-glucans, which exhibit slowly digestible properties. These alpha-glucans contain a higher proportion of branched alpha-LDx compared to other starches. When hydrolyzed by mammalian alpha-glucosidases, the glucose generation rate decreases due to the increased amount of embedded branched alpha-LDx.
