Novel fluoroquinolones containing 2-arylamino-2-oxoethyl fragment: Design, synthesis, evaluation of antibacterial and antituberculosis activities and molecular modeling studies

Summary: Novel substituted fluoroquinolone derivatives were synthesized and evaluated for their antituberculosis and antibacterial activity. Compound 14 exhibited the most potent antimicrobial activity against all tested bacteria. Most of the compounds showed antimycobacterial effects, with compounds 14 and 18 displaying the highest activity. In addition, these compounds demonstrated a similar binding mode to DNA gyrase enzymes as known fluoroquinolone drugs.

Summary: 1,3,4-Oxadiazoles and 1,2,3-oxadiazoles are important heterocyclic compounds with potential biological activities. A series of novel 1,2,3-oxadiazole derivatives with antimicrobial activities were synthesized, among which compound 5f demonstrated good inhibition against all tested strains. In silico analyses predicted that compound 5f had extensive and stable interaction with target proteins, while ADMET analysis showed no toxicity and carcinogenicity, making it a potential drug candidate for the future.

Exploring the Antimicrobial and Pharmacological Potential of NF22 as a Potent Inhibitor of E. coli DNA Gyrase: An In Vitro and In Silico Study

Summary: In the search for novel antimicrobial agents to combat pathogenic E. coli infections, a series of norfloxacin derivatives were screened. One particular derivative, NF22, showed excellent antibacterial activities against E. coli strains, including multidrug-resistant strains. NF22 was found to be more effective than norfloxacin and ciprofloxacin in killing both control and multidrug-resistant E. coli strains. Combination studies with tetracycline demonstrated a synergistic effect. Molecular modeling studies on DNA gyrase supported the experimental findings, providing insights into the interactions involved. Additionally, NF22 was evaluated for its pharmacokinetic properties and drug-likeness. The study highlights the potential of norfloxacin in restoring its efficacy against E. coli and the development of new antimicrobial agents.

Exploration of Spirocyclic Derivatives of Ciprofloxacin as Antibacterial Agents

Summary: This study reported a new derivative of ciprofloxacin and synthesized 36 new fluoroquinolone derivatives through the synthesis of previously reported and newly synthesized derivatives of 1-oxa-9-azaspiro[5.5]undecane. The antibacterial experiments showed that compared to ciprofloxacin, the new compounds had a narrower activity spectrum, but displayed significant activity against two strains: Acinetobacter baumannii 987 (R) and Bacillus cereus 138 (R).

Characteristics of Biofilms Formed by C. parapsilosis Causing an Outbreak in a Neonatal Intensive Care Unit

Summary: This study investigates the transmission route and biofilm of Candida parapsilosis in a neonatal intensive care unit (NICU). Microsatellite analysis confirms that the C. parapsilosis strains in the three inpatients and two environmental cultures are the same. Dry weight measurements and electron microscopy confirm the formation of biofilms by C. parapsilosis. The outbreak is attributed to horizontal transfer through the incubator's humidifier, and the biofilm produced by C. parapsilosis is linked to its invasive and infectious properties.

Synthesis of Novel 3-(Piperazin-1-yl)-1,2-benzothiazole Derivatives and Their Antibacterial Activity

Summary: A new library of structurally modified 3-(piperazin-1-yl)-1,2-benzothiazole derivatives has been designed and synthesized. Their structures were characterized and their drug-likeness evaluated according to Lipinski's rule of five. The synthesized compounds were also screened for antibacterial activity, with three of them showing activity. The compound with RO5 violations exhibited almost no antibacterial activity.

Antibacterial Activity of Fluorobenzoylthiosemicarbazides and Their Cyclic Analogues with 1,2,4-Triazole Scaffold
