Tau PET Imaging in Neurodegenerative Disorders

Summary: The introduction of PET ligands that bind tau pathology has allowed for the quantification and visualization of tau pathology in aging and Alzheimer's disease. While these ligands are effective in binding tau aggregates in advanced stages of AD, their binding in non-AD tauopathies is weaker and overlaps with known off-target binding regions. An increased neocortical tau PET signal holds important prognostic information, and tau PET shows great potential as a prognostic marker in symptomatic stages of AD. For differential diagnosis, tau PET is primarily useful in distinguishing AD dementia from other neurodegenerative diseases, but its performance decreases in the mild cognitive impairment stage of AD.

Long-term test-retest of cerebral [18F]MK-6240 binding and longitudinal evaluation of extracerebral tracer uptake in healthy controls and amnestic MCI patients

Elevated Monoamine Oxidase-A in Anterior Cingulate of Post-Mortem Human Parkinson's Disease: A Potential Surrogate Biomarker for Lewy Bodies?

Summary: This study evaluated the use of anti-ubiquitin immunohistochemistry for Lewy bodies and correlated it with monoamine oxidase-A using the imaging agent [F-18]FAZIN3. The findings showed the presence of Lewy bodies in all Parkinson's disease brains and increased binding of [F-18]FAZIN3 to MAO-A in PD. This suggests that [F-18]FAZIN3 could be a potential novel PET imaging approach.

Evaluation of tau deposition using 18F-PI-2620 PET in MCI and early AD subjects-a MissionAD tau sub-study

Summary: This study aimed to evaluate tau deposition using F-18-PI-2620 PET in beta-amyloid positive subjects with mild cognitive impairment (MCI) or mild AD dementia, and investigate its correlation with amyloid deposition, CSF assessment, hippocampal volume, and cognition. The results showed significant associations between tau deposition and hippocampal volume, CSF biomarkers, and amyloid-beta load. The quantifiable increases in F-18-PI-2620 SUVR in regions with early tau deposition support the hypothesis that cortical tau is associated with cognitive impairment in AD. The study highlights the utility of F-18-PI-2620 PET in assessing tau deposits in early AD cases.

Confirmation of Lewy bodies and alpha-synuclein aggregates in human PD brain sections further supported the potential of [F-18]nifene as a valuable tool for PET imaging studies of PD.
