Natural Products as Modulators of Nrf2 Signaling Pathway in Neuroprotection

Ignacio Moratilla-Rivera et al.

Summary: Neurodegenerative diseases (NDs) are common diseases affecting the West due to increased life expectancy. Cells accumulate oxidative damage, triggering neurodegeneration. The transcription factor Nrf2 regulates endogenous antioxidant systems and induces the transcription of genes containing ARE in response to prooxidant conditions. Natural products such as quercetin, curcumin, and terpenoids can positively modulate Nrf2, reducing oxidative damage to the nervous system. This review aims to update knowledge on the potential treatments for NDs by activating the Nrf2 pathway with secondary metabolites of health interest.

Multitarget Hybrid Fasudil Derivatives as a New Approach to the Potential Treatment of Amyotrophic Lateral Sclerosis

Olmo Martin-Camara et al.

Summary: Hybrid compounds containing structural fragments of the Rho kinase inhibitor fasudil and the NRF2 inducers caffeic and ferulic acids were designed, and one of the compounds showed good antioxidant properties and tolerability. This compound was able to induce the NRF2 signature and promote the expression of antioxidant response enzymes. The study suggests the therapeutic potential of this compound for ALS patients, particularly those with a SOD1 mutation.

CAPE and its synthetic derivative VP961 restore BACH1/NRF2 axis in Down Syndrome

Sara Pagnotta et al.

Summary: This study focuses on the role of BACH1/NRF2 ratio in the regulation of antioxidant response and its impact on patients with Down syndrome. The results show that overexpression of BACH1 disrupts the induction of antioxidant response genes, leading to oxidative damage accumulation. Additionally, administration of CAPE and VP961 promotes NRF2 nuclear translocation in cells from Down syndrome patients, improving antioxidant response.

Boosting caffeic acid performance as antioxidant and monoamine oxidase B/catechol-O-methyltransferase inhibitor

Daniel Chavarria et al.

Summary: CA derivatives have antioxidant properties and inhibitory effects on MAOs and COMT. They can protect cells against oxidative damage, but may induce ROS generation and cytotoxic effects at high concentrations. Some compounds can cross the blood-brain barrier, potentially aiding in restoring DA neurotransmission and preventing PD.

Extra Virgin Olive Oil Phenolic Extract on Human Hepatic HepG2 and Intestinal Caco-2 Cells: Assessment of the Antioxidant Activity and Intestinal Trans-Epithelial Transport

Martina Bartolomei et al.

Summary: This study provides a comprehensive characterization of the antioxidant properties of phenolic extract (BUO) obtained from extra virgin olive oil, cultivated in Tuscany, Italy. The findings show that BUO extract can reduce harmful substances produced in cells after oxidative stress and demonstrate selective trans-epithelial transport of some phenolic components in differentiated Caco-2 cells, a mature enterocyte model.

Monoamine oxidases in age-associated diseases: New perspectives for old enzymes

Yohan Santin et al.

Summary: Population aging leads to a higher prevalence of chronic diseases and rising healthcare costs. Cellular senescence and mitochondrial oxidative stress are key processes in age-related diseases. Monoamine oxidases (MAOs) play a role in pathological aging by being upregulated in aged or dysfunctional organs, contributing to the increased degradation of MAO substrates.

Neuroprotective and antioxidative effects of pioglitazone in brain tissue adjacent to the ischemic core are mediated by PI3K/Akt and Nrf2/ARE pathways

Yi Zhao et al.

Summary: Pioglitazone exerts neuroprotective effects by activating the PI3K/Akt pathway to inhibit apoptosis in ischemic brain tissue, as well as activating the antioxidant defense pathway through Nrf2, supporting the survival of injured neurons.

Safety and efficacy of pioglitazone for the delay of cognitive impairment in people at risk of Alzheimer's disease (TOMMORROW): a prognostic biomarker study and a phase 3, randomised, double-blind, placebo-controlled trial

Daniel K. Burns et al.

Summary: The study aimed to identify participants at risk of developing mild cognitive impairment due to Alzheimer's disease using the BRAA algorithm and evaluate the safety and efficacy of low-dose pioglitazone. Results showed that pioglitazone did not delay the onset of mild cognitive impairment.

Reassessment of Pioglitazone for Alzheimer's Disease

Ann M. Saunders et al.

Summary: Alzheimer's disease is a significant unmet medical need in the elderly population, with no treatments available to reduce the risk of developing AD or to offer prolonged mitigation of its devastating symptoms. Despite promising results in pre-clinical experiments and observational studies, placebo-controlled clinical trials have not shown the same efficacy for pioglitazone in delaying the onset of dementia in individuals with pre-existing type 2 diabetes mellitus.
