Phospholipase Cγ1 (PLCG1) overexpression is associated with tumor growth and poor survival in IDH wild-type lower-grade gliomas in adult patients

Tianwen Li et al.

Summary: In IDH wild-type lower-grade gliomas, the mRNA expression levels of phospholipase C gamma 1 (PLCG1) are much higher compared to IDH mutant LGGs, indicating a correlation between elevated PLCG1 expression and poor clinical outcome. PLCG1 amplification may act as a key mechanism for upregulation. Depletion of PLCG1 expression can inhibit proliferation and invasion of IDH wild-type LGG cell lines.

Impact of phospholipase C β1 in glioblastoma: a study on the main mechanisms of tumor aggressiveness

Stefano Ratti et al.

Summary: This study aims to determine the pathological impact of PLC beta 1 in glioblastoma. The results showed that PLC beta 1 gene expression correlates with glioma's grade, and silencing PLC beta 1 leads to increased cell migration, invasion, and proliferation in glioblastoma cells.

Recurrent Glioblastoma: From Molecular Landscape to New Treatment Perspectives

Cristina Birzu et al.

Summary: Glioblastoma is the most common and aggressive primary brain cancer in adults. Despite standard treatments including surgery, radiotherapy, and chemotherapy, prognosis remains poor with relapse being common. New treatment options such as immunotherapy and targeted therapy have shown promise in recent years for recurrent cases.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Development of 2-Morpholino-N-hydroxybenzamides as anti-proliferative PC-PLC inhibitors

Shaun W. P. Rees et al.

Summary: PC-PLC enzyme is a potential target for anti-cancer treatment due to its involvement in cellular processes related to oncogenesis. Modification of the carboxylic acid motif on 2-morpholinobenzoic acids with hydroxamic acid enhances the inhibitory activity of PC-PLC and shows promising anti-proliferative effects in cancer cell lines.

An optimised MALDI-TOF assay for phosphatidylcholine-specific phospholipase C

Nabangshu Sharma et al.

Summary: The study reports the development and optimisation of a MALDI-TOF mass spectrometry-based assay to monitor PC-PLCBc activity, comparing the inhibition of PC-PLCBc by different structural classes of inhibitors in one-phase and two-phase reaction systems. The assay was shown to have advantages over the commonly used Amplex Red assay and can be applied to study the activity and inhibition of other phospholipases.
