Screening of Zwitterionic Liposomes with Red Blood Cell-Hitchhiking and Tumor Cell-Active Transporting Capability for Efficient Tumor Entrance

Summary: A type of tertiary amine oxide (TAO)-containing zwitterionic liposomal nanocarriers has been developed to hitchhike red blood cells (RBCs) to tumor blood vessels and enter solid tumors through transcytosis. Two types of TAO liposomes (TAOLs) have been identified that can induce efficient tumor cell transcytosis through rapid macropinocytosis and endoplasmic reticulum/Golgi-involved exocytosis. These zwitterionic TAOLs can hitchhike RBCs to gain long blood circulation and effectively enter and infiltrate the whole tumor through transcytosis. Chemotherapeutic drug-loaded liposomes have shown superior antitumor activity compared to traditional liposomal drugs.

The pH-responsive dissolution properties of the PAA-CaP passivation layer resulted in faster cumulative DOX release at acidic pH, leading to higher cytotoxicity towards MCF-7 tumor cells. The enhanced cellular uptake and intracellular release of DOX contributed to the higher total cytotoxicity of TiO2@PAA-CaP(DOX) NPs under UV-A irradiation due to the synergistic effect between efficient DOX delivery and enhanced photoinduced reactive oxygen species (ROS) generation.

Vascular bursts-mediated tumor accumulation and deep penetration of spherical nucleic acids for synergistic radio-immunotherapy

Summary: A two-step low-dose radiotherapy strategy combined with spherical nucleic acids-based nanomedicine was used to enhance the accumulation and penetration of nanoparticles in tumors. The strategy successfully recruited macrophages into the tumor, which not only served as drug depots but also enhanced extravasation to improve nanoparticle accumulation. Through optimized timing and administration, the delivered nanoparticles promoted a phenotype switch in macrophages, resulting in increased phagocytosis and further enhancing accumulation and penetration. This approach showed significant enhancement of nanoparticles accumulation compared to passive targeting, with efficient deep penetration throughout the tumor tissue.

Nanozymes-engineered metal-organic frameworks for enhanced microwave thermodynamic therapy in PDX of hepatic carcinoma
