FOSL1 promotes proneural-to-mesenchymal transition of glioblastoma stem cells via UBC9/CYLD/NF-KB axis

Summary: FOSL1 is a key player in regulating the proneural to mesenchymal transition (PMT) in glioblastoma stem cells (GSCs). It promotes the ionizing radiation-induced PMT and radioresistance of Proneural GSCs. Mechanistically, FOSL1 induces PMT by promoting UBC9-dependent CYLD SUMOylation and subsequent NF-kappa B activation. Therapeutic targeting of FOSL1 holds promise to attenuate molecular subtype switching in glioblastoma patients.

Natural Coevolution of Tumor and Immunoenvironment in Glioblastoma

The researchers identify a natural evolution signature (NES) in the tumor and find that it is associated with the activation of transcription factors involved in brain development. A RUNX-targeted gene switch-off approach modulates the BIRC5/PIF1-p21 pathway and reduces glioblastoma growth in mice

Summary: Interfering with RUNX family proteins can reduce the growth of glioblastoma in mice and identify pathways involved in tumor growth maintenance. Specifically, the study reveals that RUNX1, a transcription factor, downregulates p21 by enhancing the expressions of BIRC5 and PIF1, thus conferring anti-apoptotic properties on glioblastoma. Gene switch-off therapy targeting RUNX1 shows promising results by decreasing the expression levels of BIRC5 and PIF1, inducing apoptosis and cell cycle arrest. This pathway may serve as a therapeutic target for glioblastoma.

Epigenetic encoding, heritability and plasticity of glioma transcriptional cell states

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.

Integrated analysis of methylome and transcriptome data from MLLr and non-MLLr iB-ALL patients revealed chromatin state alterations and identified distinct gene expression patterns, particularly related to the AP-1 complex and RUNX factors in MLLr iB-ALL. Pharmacological inhibition of AP-1 showed promising potential in impairing MLLr-leukemic growth, suggesting a novel therapeutic approach for MLLr-iB-ALL.
