Dynamic phase separation of the androgen receptor and its coactivators key to regulate gene expression

Summary: Numerous cancers, including prostate cancer (PCa), rely on transcription programs driven by specific genomic regions called super-enhancers. The androgen receptor (AR), the main oncogenic driver in PCa, forms liquid-like foci in different PCa models, and this foci formation is correlated with AR transcriptional activity. AR antagonists inhibit foci formation and phase separation of AR, suggesting that enhanced compartmentalization of AR and coactivators may play a crucial role in the activation of oncogenic transcription programs in androgen-dependent PCa.

Phase separation of RNA-binding protein promotes polymerase binding and transcription

Summary: A new study suggests that RNA-binding proteins constitute half of the chromatin proteome in embryonic stem cells, with some colocalizing with RNA polymerase II at promoters and enhancers. These RNA-binding proteins inhibit premature release of Pol II and enhance the formation of transcription condensates using RNA as multivalent molecules, ultimately promoting polymerase engagement and activity. The study proposes a synergistic interplay between promoter-associated RNAs and their binding proteins to facilitate active transcription through phase separation of polymerase condensates.

Transcription activation is enhanced by multivalent interactions independent of phase separation

Summary: This study investigates the transcriptional activation process by comparing synthetic transcription factors (TFs) at a reporter gene array. The results show that the physicochemical properties of the activation domain (AD) can control TF assembly at chromatin. It was found that while the propensity for phase separation and activation strength of the AD were correlated, the actual formation of liquid-like TF droplets had a neutral or inhibitory effect on transcription activation. The study concludes that multivalent AD-mediated interactions enhance transcription activation capacity by increasing the TF's residence time in the chromatin-bound state and facilitating the recruitment of coactivators, independent of phase separation.
