Cancer Statistics, 2021

Rebecca L. Siegel et al.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Comparing programmed death ligand 1 scores for predicting pembrolizumab efficacy in head and neck cancer

Kenneth Emancipator et al.

Summary: Tumor proportion score (TPS) and combined positive score (CPS) are two methods used to evaluate PD-L1 expression, and CPS >= 50 appears to be equivalent to TPS >= 50% in predicting response rates. CPS may be more sensitive than TPS at lower cutoffs.

Effects of checkpoint kinase 1 inhibition by prexasertib on the tumor immune microenvironment of head and neck squamous cell carcinoma

Ritu Chaudhary et al.

Summary: Prexasertib shows potential therapeutic effects in HNSCC, but long-term resistance may be related to evasion of immune surveillance. Genetics and immunohistochemistry analyses suggest significant impact of Prexasertib on the tumor immune microenvironment.

Defining HPV-specific B cell responses in patients with head and neck cancer

Andreas Wieland et al.

Summary: Detailed analyses of B cells in the tumor microenvironment of HPV-linked head and neck cancers reveal strong humoral immune responses to HPV antigens and the secretion of HPV-specific antibodies in situ. The findings suggest that tumor-infiltrating B cells could potentially be utilized for the development of therapeutic agents in human cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

PD-L1 as a biomarker of response to immune-checkpoint inhibitors

Deborah Blythe Doroshow et al.

Summary: PD-L1 expression is currently the best predictor of responsiveness to immune-checkpoint inhibitors, despite challenges in clinical use such as variability in different assays and cut-off points, as well as the lack of prospective comparisons with clinical outcomes. Nevertheless, several immunohistochemical assays have been approved for clinical use, with implications for pathology services and potential impacts on clinical outcomes. Comparison of FDA-approved PD-L1 assays, understanding of the varying implications of PD-L1 expression across different tumour types, and exploration of novel approaches to optimize its clinical utility are important topics for further research and practice.

TIMEx: tumor-immune microenvironment deconvolution web-portal for bulk transcriptomics using pan-cancer scRNA-seq signatures

Mengyu Xie et al.

Summary: Heterogeneous cell types within the tumor-immune microenvironment play vital roles in cancer progression, metastasis, and treatment response. TIMEx, a novel deconvolution method, focuses on estimating infiltrating immune cells using pan-cancer single-cell RNA-seq signatures. Additionally, a user-friendly web portal has been developed to allow users to evaluate TIMEx and other deconvolution methods with bulk transcriptomic profiles.

Tumor microenvironment and immune evasion in head and neck squamous cell carcinoma

Areeg Elmusrati et al.

Summary: Head and neck squamous cell carcinoma (HNSCC) is an aggressive malignancy characterized by high morbidity and low survival rates, with limited therapeutic options. The immune system plays a key role in cancer surveillance against HNSCC initiation and progression. The interaction between cancer cells and the host immune system has a detrimental effect on the tumor microenvironment, promoting angiogenesis, proliferation, and metastasis.

B cell signatures and tertiary lymphoid structures contribute to outcome in head and neck squamous cell carcinoma

Ayana T. Ruffin et al.

Summary: The study suggests that enhancing TIL-B responses in HNSCC, particularly in HPV+ cases, could complement current T cell immunotherapies. The presence of tumor infiltrating B cells with GCs in TLS is associated with better survival outcomes in head and neck squamous cell carcinoma patients.
