Development and evaluation of pulsed - Post column derivatization in liquid chromatography as a concept to minimize reagent consumption

Summary: In this article, a new approach to reduce the consumption of reagents in liquid chromatography coupled to online post column derivatization is proposed. The proposed method, called Pulsed-Post Column Derivatization (Pulsed-PCD), introduces reagents as well-defined pulses at microliter levels. The robustness of the procedure was evaluated and the method was successfully applied to determine histidine in human urine.

Pulsed-post column derivatization coupled to green liquid chromatography for the determination of glutathione and cysteine based on thioacrylates formation

Summary: In this study, a new method called Pulsed-post column derivatization (Pulsed-PCD) was developed for the determination of thiols in yeast samples. The method involved using 100% aqueous mobile phase for chromatographic separation and on-line reaction with a derivatizing reagent. Spectrophotometric detection was carried out and the method was validated for endogenous content determination. The proposed method was found to be greener in terms of chemical usage and waste production compared to a corroborative method based on classical PCD.

Development and validation of a direct HPLC method for the determination of salivary glutathione disulphide using a core shell column and post column derivatization with o-phthalaldehyde

Summary: In this study, an optimized and validated method was used to monitor glutathione disulfide (GSSG) in human saliva samples. The results showed accurate quantification of GSSG in the samples, with a concentration range of 100-2000 nmol L-1 and a LOD of 20 nmol L-1. The stability of the samples was ensured by blocking reduced glutathione with N-ethylmaleimide.

CF patients exhibit an abnormal proinflammatory environment in their airways even before infection, possibly due to elevated oxidative stress and abnormal lipid metabolism. CFTR deficiency appears to cause a redox imbalance in epithelial cells and extracellular fluids.
