TonB-dependent receptor epitopes expressed in M. bovis BCG induced significant protection in the hamster model of leptospirosis

Everton B. Bettin et al.

Summary: Leptospirosis is an emerging infectious disease caused by pathogenic Leptospira spp. Developing a universal vaccine requires identifying highly conserved epitopes exposed on the bacterial surface. A recombinant BCG vaccine targeting leptospiral TBDRs induced a protective and sterilizing immune response in Syrian hamsters, showing promise for preventing leptospirosis.

Summary: This study investigates the role of leptospiral protein antigens in the activation of WC1(+) γδ T cells and identifies two leptospiral outer membrane proteins that can directly interact with them. Protein-specific γδ T cell responses consist of WC1.1(+) and WC1.2(+) subsets.

Summary: This study utilizes CRISPR interference technology to perform gene silencing in Leptospira, and identifies differentially expressed proteins in mutant strains compared to the control. The study also investigates the impact on virulence of the mutant strains and provides insights into the pathogenic mechanisms of Leptospira.

Challenges for the development of a universal vaccine against leptospirosis revealed by the evaluation of 22 vaccine candidates

Mara A. C. Maia et al.

Summary: Leptospirosis is a neglected disease that affects humans and animals. Current vaccines fail to induce cross-protective immune response. Researchers attempted to design a new vaccine by identifying conserved epitopes of leptospiral antigens, and tested it in a hamster model. The vaccine showed some protective effect, but the results were not reproducible in subsequent experiments. Further evaluation of alternative animal models is needed for leptospirosis vaccine research.

The Diverse Applications of Recombinant BCG-Based Vaccines to Target Infectious Diseases Other Than Tuberculosis: An Overview

Esma Mouhoub et al.

Summary: Live attenuated Bacillus Calmette-Guerin (BCG) vaccine is widely used for preventing tuberculosis, but has also shown protection against other pathogens. Recombinant BCG technology has been used to create vaccines that elicit immunity against bacterial, viral, and parasitic diseases. This review aims to provide an overview of rBCG-based vaccines targeting infectious diseases other than TB.
