Complex regulation in a Comamonas platform for diverse aromatic carbon metabolism

Rebecca A. Wilkes et al.

Summary: Microbial platforms are crucial for a sustainable energy future and can process aromatic carbons from lignin and plastic feedstocks. Comamonas species are promising bacterial candidates due to their ability to use natural and xenobiotic aromatic compounds, and they often have genetic constraints that prevent competition with sugars. However, the metabolic reactions of these species and their regulatory mechanisms are not well understood.

High-level production of 2-pyrone-4,6-dicarboxylic acid from vanillic acid as a lignin-related aromatic compound by metabolically engineered fermentation to realize industrial valorization processes of lignin

Yuichiro Otsuka et al.

Summary: This study aimed to design a fermentation process for producing 2-pyrone-4,6-dicarboxylic acid (PDC) from vanillic acid, which could be applied to industrial production. Metabolomic analysis revealed that a high primary metabolic activity within cells was required to improve production efficiency. The resulting optimized process yielded the highest reported PDC titer and productivity: 99.9 g/L and 1.69 g/L/h, respectively.

Recent advances and challenges in the biotechnological upcycling of plastic wastes for constructing a circular bioeconomy

Sangmin Lee et al.

Summary: Plastic waste upcycling has emerged as a solution to the environmental crisis caused by plastic, with a focus on enzymatic and microbial degradation as well as catabolism of plastic monomers. This review provides insights into recent advances and barriers in biotechnological upcycling, specifically discussing PETase as a representative plastic-degrading enzyme. It also covers microbial conversion of plastic monomers and the chemobiological upcycling of plastic waste, contributing to the development of viable biotechnological systems and plastic-based circular carbon economies.

Advanced PHB fermentation strategies with CO2-derived organic acids

Elodie Vlaeminck et al.

Summary: Recent research has shown that acetic acid is more suitable as a carbon source for PHB production compared to formic acid, possibly due to differences in their metabolism and utilization during fermentation. By optimizing fermentation conditions and introducing new strategies, a highly efficient and stable process for PHB production was successfully established.

Towards bio-upcycling of polyethylene terephthalate

Till Tiso et al.

Summary: With over 359 million tons of plastics produced worldwide in 2018, the challenge of managing plastic waste is evident. Recent discovery of enzymes that degrade supposedly non-biodegradable plastics has opened up opportunities for transforming the plastic recycling industry using biotechnology. This study presents a novel method for converting post-consumer PET into bioplastics, offering a solution that bypasses the expensive purification process for PET monomers and adds technological flexibility to the global issue of plastic waste management.
