Summary: In this study, multifunctional stimuliresponsive metal-organic framework (MOF) hydrogels were constructed on the surface of the sulfonated long carbon fiber reinforced polyetheretherketone (LCFRPEEK) implant to coordinate with the osseointegration process. The hydrogels composed of HAP@Mg-GA MOF nanoparticles and methacryloyl chitosan demonstrated outstanding pH-sensitive biomolecule release capabilities. The in vitro and in vivo experiments confirmed the immunomodulation, angiogenesis, osteogenic differentiation, and osseointegration abilities of the implant.

The Role of Surface Chemistry in the Osseointegration of PEEK Implants

Summary: Poly(etheretherketone) (PEEK) implants with added NH2 and COOH groups on their surface can modulate macrophage responses and improve osseointegration. NH2-functionalized surfaces induce pro-inflammatory macrophage responses, while COOH-functionalized surfaces induce anti-inflammatory macrophage responses. The protein adsorption patterns on these surfaces are related to the varied inflammatory responses. NH2 surfaces inhibit the osteogenic differentiation of mesenchymal stem cells (MSCs) compared to COOH surfaces, despite promoting more mineralization in simulated body fluid solutions. In the rat tibia unicortical defects study, the bone around NH2 surfaces had thinner trabeculae and higher specific bone surface, indicating improved bone binding compared to unmodified implants. COOH surfaces only showed a trend in increasing bone binding. The results suggest that a combination of both mineral-binding and immune responses can improve osseointegration of PEEK implants.

Summary: Implant-associated infection and insufficient implant osseointegration are common causes of implant failure. In this study, a multi-functional bioactive ion, copper (Cu), was incorporated into the polydopamine (PDA) coating on polyetheretherketone (PEEK) surfaces. The DA-Cu-II coated PEEK surfaces showed superior abilities of osteogenesis, angiogenesis, and antibiosis, as well as immunomodulatory effects. The coating also induced a mild and necessary inflammatory response, exhibiting excellent antibacterial activity and osseointegration. The findings suggest that the DA-Cu-II coated PEEK has great potential for orthopedic implants and provides new insights into novel modification strategies for PEEK materials.

The Single-Step Fabrication of a Poly (Sodium Vinylsulfonate)-Grafted Polyetheretherketone Surface to Ameliorate Its Osteogenic Activity

Summary: Polyetheretherketone (PEEK) is a potential material for orthopedic implants due to its similar elastic modulus to human bone. However, its poor osteogenic activity limits its clinical application. In this study, PEEK surface was grafted with poly (sodium vinylsulfonate) to improve its osteogenic activity. The results showed that grafting with poly (sodium vinylsulfonate) enhanced MC3T3-E1 osteoblast compatibility and osteogenic activity of the PEEK surface, especially with a higher grafting amount.

Enhancing antibacterial capability and osseointegration of polyetheretherketone (PEEK) implants by dual-functional surface modification
