Enhanced phycocyanin production from Spirulina subsalsa via freshwater and marine cultivation with optimized light source and temperature

Summary: To determine the best strategy to produce phycocyanin in a cost-efficient way, the impact of light source and temperature on the growth of Spirulina subsalsa was examined. The results revealed that the highest growth rate was achieved at 35 degrees C, while the highest phycocyanin content was obtained under green light. Furthermore, a two-stage cultivation strategy combining biomass accumulation at 35 degrees C and phycocyanin synthesis under green light was proposed and implemented, resulting in significant phycocyanin production.

Fluorescence and antioxidant activity of heterologous expression of phycocyanin and allophycocyanin from Arthrospira platensis

Summary: In this study, recombinant expression was used to produce different phycocyanin and allophycocyanin variants. The molecular weights of these recombinant proteins were identified, and their fluorescent and antioxidant activities were analyzed. The results showed that the recombinant phycocyanin-allophycocyanin polymer had stronger antioxidant activity, while recombinant phycocyanin had stronger DPPH antioxidant activity.

Efficient Synthesis of Phycocyanobilin by Combinatorial Metabolic Engineering in Escherichia coli

Summary: This study reports the efficient synthesis of Phycocyanobilin (PCB) in Escherichia coli by expressing heme oxygenase-1 from Thermosynechococcus elongatus and PCB:ferredoxin oxidoreductase from Synechocystis sp. The synthesis of PCB was further enhanced by direct supplementation of 5-aminolevulinic acid (ALA), moderate overexpression of key enzymes in the heme biosynthetic pathway, and accelerated cycle of cofactors. The highest reported titer of PCB (28.32 mg/L) was obtained under optimal conditions using a fermenter.

This study identified 84 gene targets through computational simulations, of which 40 were confirmed to enhance heme production in experiments. Not only genes directly involved in heme biosynthesis, but also genes related to glycolysis, glycine, and succinyl-coenzyme A metabolism were found to affect heme production.

PCC6803 were investigated. Knocking out spkD and spkG genes led to lower levels of certain fatty acids and altered expression of fatty acid desaturase genes, indicating their importance in polyunsaturated fatty acid biosynthesis and potential for genetic engineering in cyanobacteria.
