Reagents for Mass Cytometry

Loryn P. Arnett et al.

Summary: Mass cytometry (CyTOF) is a bioanalytical technique that allows for identification and quantification of cellular features at single-cell resolution. It utilizes stable heavy-atom isotope-tagged reagents for analysis in suspension or imaging mass cytometry. The development of new reagents, such as small molecule reagents, has expanded the scope of information obtainable. This review provides a summary of the history and current state of mass cytometry reagent development, as well as guidelines for testing and presentation of new reagents.

Either IL-7 activation of JAK-STAT or BEZ inhibition of PI3K-AKT-mTOR pathways dominates the single-cell phosphosignature of ex vivo treated pediatric T-cell acute lymphoblastic leukemia cells

Daniela Kuzilkova et al.

Summary: This study utilized single-cell mass cytometry to investigate signal transduction pathways in T-cell acute lymphoblastic leukemia (T-ALL), and found that different cellular clusters exhibited varying responsiveness to therapeutic drugs, which remained consistent during relapse.

Standardization of Workflow and Flow Cytometry Panels for Quantitative Expression Profiling of Surface Antigens on Blood Leukocyte Subsets: An HCDM CDMaps Initiative

Daniela Kuzilkova et al.

Summary: This study developed a flow cytometric procedure for standardized quantitative expression profiling of surface antigens on blood leukocyte subsets across multiple research laboratories. The designed tubes can discriminate 27 leukocyte subsets and detect the expression of PE-conjugated CD markers.

HPMA Copolymer-Based Nanomedicines in Controlled Drug Delivery

Petr Chytil et al.

Summary: Water-soluble polymer carriers, covalently attached to low-molecular-weight drugs using biodegradable spacers, are important in drug delivery systems. Synthetic polymer biomaterials based on HPMA copolymers are recognized as serious candidates for nanomedicine due to their unique properties. The main impact of HPMA polymer-based drug carriers is the improvement of targeting ability, especially through the EPR effect.

The role of the biotin linker in polymer antibody mimetics, iBodies, in biochemical assays

Vladimir Subr et al.

Summary: The study focused on the impact of the linker between the polymer backbone and biotin, the number of biotins per polymer chain in ELISA or pull-down assays, and the position of biotin on the polymer chain. A novel chain transfer agent suitable for RAFT polymerization was developed to functionalize polymer precursors and simplify the synthesis of semitelechelic antibody mimetic materials. Optimized iBodies with a lengthened linker between the polymer backbone and biotin increased ELISA sensitivity up to 5 times.
