Lactate-induced lactylation in skeletal muscle is associated with insulin resistance in humans

Summary: This study found that lactate-induced lysine lactylation is present in human skeletal muscle, and its levels are increased in obese individuals and associated with insulin resistance. Skeletal muscle lactylation is related to anaerobic metabolism and negatively correlated with oxidative metabolism. In in vitro experiments, treating human skeletal muscle cells with lactate increased protein lactylation, and inhibiting glycolysis or LDH-A reduced lactylation levels. This study suggests that skeletal muscle lactylation could provide additional insight into the regulation of skeletal muscle metabolism, including insulin resistance.
